Aim:With the guidance of the theory of traditional Chinese medicine,to observe the effect of traditional Chinese medicine(TCM) compound Anshen Decoction on conditioned place preference (CPP), behavioral sensitization and the concentration of neurotransmitter dopamine (DA),glutamic acid (Glu) and y-aminobutyric acid(GABA) in the mesolimbic region of alcohol addiction mice, and to explore its intervention mechanism of alcohol addiction. In order to provide theoretical basis and new treatment ideas for the prevention and treatment of alcohol addiction.Method:1.48 SPF grade mice, male, weighting 22±3g,were randomly divided into four groups, that was saline group, alcohol group, Chinese medicine group and Chinese medicine intervention group, each group of 12. Chinese herbs group and Chinese herbs intervention group were given Anshen Decoction, saline group, alcohol group were given equal volume of normal saline.30 minutes later,in addition to saline group and Chinese medicine group, the other groups were given alcohol to establish CPP model.To evaluate the effect of Anshen Decoction on CPP induced by alcohol and whether or not it is addictive, through by the residence time in cabinet with medicine before and after the experiment.2.160 SPF grade mice, male, weighting 22±3g,were randomly divided into four groups, that was saline group, alcohol group, Chinese medicine group and Chinese medicine intervention group, each group of 40. Chinese herbs group and Chinese herbs intervention group were given Anshen Decoction, saline group, alcohol group were given equal volume of normal saline.30 minutes later,in addition to saline group and Chinese medicine group, the other groups were given alcohol to establish and behavioral sensitization model.To evaluate the effect of Anshen Decoction on the development and expression of behavioral sensitization induced by alcohol and its own effect on spontaneous activity of mice, through by observing the spontaneous activity in behavioral sensitization experiment.3.40 SPF grade mice, male, weighting 22±3g,were randomly divided into saline group,alcohol group, development of behavioral sensitization intervention group and expression of behavioral sensitization intervention group,each group of 10, to establish and behavioral sensitization model.The mice were decapitated 15 minutes after giving alcohol, or Anshen Decoction and alcohol.The enzyme linked immunosorbent assay (ELISA) was used to determine the concentration of DA, Glu and GABA in mesolimbic area of mice.Results:1. The residence time in the medicine cabinet of Chinese medicine group mice was 239.0±48.50s, there is no significant difference compared with saline group (p>0.05). The residence time in the medicine cabinet of Chinese medicine intervention group was 298.7±59.50s, obviously lower than alcohol group, and there is significant difference (p< 0.01).2. On 1,3,5,7, and 9 day,compared Chinese medicine intervention group with alcohol group,the locomotor activity times of Chinese medicine group were 238.9+ 51.53.232.8±91.34,259.3±98.96.317.2±86.46,324.5±97.25 respectively, significantly lower than the alcohol group, and all have a statistics difference (p< 0.05 or p< 0.01).3. On 1,3,5,7, and 9 day,the locomotor activity times of Chinese medicine group were 21.44±47.68,242.8±55.28,24.72±56.59,223.1±63.78,216.9± 62.07 respectively, and there are no statistical significance respectively compared with saline group(p> 0.05).4. The 13th day,compared Chinese medicine group challenged by Chinese medicine with saline group challenged by Chinese medicine, the locomotor activity (224.7±45.04 times), has no statistics difference (p> 0.05).5. The 13th day, compared Chinese medicine intervention group challenged by alcohol with alcohol group challenged by alcohol, the locomotor activity (386.4± 103.48 times) significantly lower than the number of the alcohol group challenged by alcohol, and has a statistics difference (p< 0.01).6. The 13th day, compared alcohol group challenged by Chinese medicine+ alcohol with alcohol group challenged by alcohol, the locomotor activity (376.0± 58.90 times) has a statistics difference (p< 0.01).7. Compared development of behavioral sensitization intervention group with model group, the DA neurotransmitter concentration(54.4±4.82ng/L), Glu neurotransmitter concentration(127.4±8.25 nmol/L) and GABA neurotransmitter concentration 13.8±1.88 ng/ml), all has a statistics difference (p< 0.05).8. Compared expression of behavioral sensitization intervention group with model group, the DA neurotransmitter concentration(58.6±5.75ng/L), Glu neurotransmitter concentration(125.8±6.48 nmol/L) and GABA neurotransmitter concentration(12.5±1.64 ng/ml), all has a statistics difference (p< 0.05).Conclusions:1. Anshen Decoction tself does not have the potential ability to lead to formation of CPP in mice, but could inhibit the formation of CPP induced by alcohol in mice.2. Anshen Decoction used alone does not affect the locomotor activity of mice, but can inhibit the high locomotor activity of mice induced by a single acute alcohol use, and also can inhibit the development and expression of behavioral sensitization induced by alcohol in mice.3. Anshen Decoction can decrease the content of DA and Glu, but increase GABA in mesolimbic areas of alcohol sensitized mice.4. Anshen Decoction itself does not have the potential ability to cause addiction, and the treatment of alcohol dependence effect is clear, can be considered as the mother liquor of Chinese herbal medicine beveraging promotion in the treatment of alcohol addiction.
