Objective:At present, clinical medicine simulation of sepsis has a lot of kinds of methods, various models have been developed. But the best animal model is the model of the ligation and perforation of the rodent's appendix. The most important method is to control the mortality rate by the thickness of the puncture needle, the number of perforation and the length of the ligation of the appendix. In this model, the key of sepsis is caused by intra-abdominal infectious microorganisms trigger systemic inflammatory response. This model has a high consistency and can be obtained by repeated experiments. The aim of this study was to establish a CLP model to further control the degree of infection and the mortality of the model, which is based on the length of the volume of the book, and the number of perforation, at the same time, according to the contrast of the previous literature, to explore the feasibility of improving the stability of the model.Methods:SD rats were performed using the male rats for the ligation and puncture. The length of the ligation of the appendix was divided into 2 groups, Measurement of the length of the total length of the top of the appendix to the end of the appendix with silk thread. Divided into 100% length and the length of the ligation group and 50%. And to observe a week of mortality.Results:The 7 day rate of 50% in the operation group was 55.6%?100% 7 day mortality was 63.9%, There was no significant difference between the 2 groups?The mortality of each subgroup increased with the increase of the number of perforation, The mortality rate was 7 at 20.8% days in the perforation group one, the perforation group two was58.3%, the perforation group three was100%, The sham operation group were all alive. The average survival time of the operation group 50% was 49.75+5.18 hours,100%was43.19±5.13hours. The average survival time of the perforation group one was 74.83+3.66 hours, perforation group two was 47.29±6.38 hours, perforation group three was 17.29±0.45 hours.Conclusions:The experimental results showed that the mortality of the group was punctured by the ligation of the appendix, In the process of sepsis, especially when the production of CLP model, To make the experimental personnel effective according to the needs of the mortality rate to produce the corresponding model.
