Objective:To observe the curative effect of jiajiansanjiasan on hemorheology and the expression of iNOS and ET-1 in rats with Hepatic Fibrosis, to discuss the mechanism and curative of jiajiansanjiasan on the treatment of Hepatic fibrosis.Methods:24 Wistar rats were randomly divided into control groups,model groups,positive control groups and Jiajiansanjiasan groups. After 7 days common breeding,all the rats were duplicated immuno-hepatic fibrosis model by injecting porcine serum intraperitoneally except the control groups. After modeling, all the rats were taken medicine by gavage, once daily.The medicine of Jiajiansanjiasan groups were taken jiajiansanjiasan powder, positive control groups were taken fufangbiejiaruanganpian powder,control groups and model groups were taken with sovolumetric 0.9%NaCl. After 60 days remedy, taking rats liver, dyeing by HE and MASSON, observing the content of coagulation indicator changes(PT,APTT,TT and FIB), testing hemodynamic indexes by SA-6000 automatic blood rheomete, exploring the contents of endothlin-1(ET-1) and Inducible Nitric Oxide Synthase (iNOS) in immunohistochemistry method, mRNA level of ET-1 and iNOS was detected by RT-PCR method.Results:1.The control group rats'hepatic lobule structure were integrated, without liver cell degeneration or necrosis, without fibrous connective tissue hyperplasia. The model group's rats appeared cellular swelling, ballooning degeneration, lipoid degeneratiaon, large scale of fibrous tissue proliferation from central veins to portal area, formation of pseudolobuli.2. Compared with control group, PT, APTT, TT in model group and positive control group and Jiajiansanjiasan group rats increased significantly; Compared with model group, PT, APTT, TT in positive control group and Jiajiansanjiasan group rats decreased significantly.FIB decreased insignificantly.3.Compared with control group, blood viscosity(200/S?5/S)?plasma viscosity,red blood cell aggregation number?HCT?erythrocyte sedimentation rate (ESR) inmodel group?positive control group and Jiajiansanjiasan group rats increased significantly. Compared with model group, blood viscosity(200/S?5/S)?HCT?erythrocyte sedimentation rate (ESR)inpositive control group and Jiajiansanjiasan group rats increased obviously.4. Compared with control group, the contents of ET-1 and iNOS increased significantly in model group;Compared with model group, ET-1 and iNOS decreased in Jiajiansanjiasan group.5. Compared with control group, mRNA level of ET-1 and iNOS of model group rats increased significantly, mRNA level of ET-1 and iNOS in Jiajiansanjiasan group and positive control group rats decreased significantly.Conclusion:1.The sanjiasan can reduce the content of coagulation indicator such as PT,APTT, TT and FIB of hepatic fibrosis rats, and it has the function of treating hepatic fibrosis rats.2.The whole blood viscosity, plasma viscosity,red blood cell aggregation number, HCT and ESR of the sanjiasan group increased than model group, which has shown sanjiasan can improve hemorheology and microcirculation of rats with immuno-hepatic fibrosis, it may be the mechanism of its anti-hepatic fibrosis function.3.The sanjiasan can down the content of vasoactive substance such as Endothelin-1, Inducible Nitric Oxide Synthase of hepatic fibrosis rats, and it has the function of treating hepatic fibrosis rats.4.The mRNA level of ET-1 and iNOS of sanjiasan group was decreased than the model group, which has shown sanjiasan can restrain vasoactive substance content of hepatic fibrosis rats. The mechanism may be it can promote the micro-circulation and release anaerobic condition to affect the expression of mRNA level of ET-1 and iNOS.
