| In order to accumulate enough of individual' ginsenosides for hemolysis test, 7 compounds were isolated and characterized as Ginsenoside-Rd(1), -Re(2), Quinquefoloside-La(3), Fidusoside-F1(4), Ginsenoside-M6a(5), Majoroside-F1(6). Among these, Quinquefoloside-La(3) is a new compound, Fidusoside-F1(4) is isolated firstly in this plant.To study the hemolytic and anti-hemolytic action of individual ginsenosides, we carry on the experiments of hemolytic action or anti-hemolytic action. Measure the absorbance of several samples which consist of different concentration of individual ginsenosides and 1% erythrocyte solution, some also contain Saponin(SIGMA), set up the simulation-relationship-curve between the concentration of individual ginsenosides and the hemolytic action or the anti-hemolytic action by statistical software, give 50% hemolysis (HD50) and 50% anti-hemolysis (P50) through data analysis. The individual ginsenosides measured in the experiment have the properties of hemolytic action and anti-hemolytic action respectively or both. Their abilities of their hemolysis or anti-hemolysis are different from each other. Protopanaxtriol-type ginsenoside-Rg1, -Re showed anti-hemolytic action, whereas -20(S)-Rg2, -20(R)-Rg2 and -Rh1 exhibited both action in different concentration; Protopanaxadiol-type ginsenoside-Rb1, -Rb2, -Re and Rd showed anti-hemolytic action, however Rd also showed hemolytic action in a higher concentration solution; Oleamolic acid type ginsenoside-Ro exhibited anti-hemolytic action only.According to the different characteristic of individual ginsenosides to the hemolytic active, we choose two of them which have hemolytic action and anti- hemolytic action that mix together and carry on anti- hemolytic test against Rd. Although both Rc and Rg1 show anti-hemolytic action against Saponin and the abilities raise up while their concentration increase, they showed different action from the experiment against Rd. The mixture of two individual ginsenosides show different action from their own one, in some situation may be the contrary characteristic.Based on the hemolytic and anti- hemolytic action of individual ginsenosides, we take Xuesaitong zhusheye witch mostly contains total Radix Notoginseng saponin for example to make a test to study the hemolytic action of different content of Xuesaitong zhusheye components by isolating and re-composing via in vitro test. The experiment data analysis by SPSS 13.0 and give out the relative regression equation: Y=-14.726+0.16X1+0.013X2-0.212X3-0.001X4-0.059X5. The equation can work out the relationship between the hemolytic action and the content composition of Xuesaitong zhusheye. |
