The Relationship Between The Depression And The Expression Of P11 MRNA In PBMC Of Hepatitis B Patients Treated With INF-?

BackgroundAlpha interferon (IFN-alpha) has the effects such as antivirus, antitumor, etc, it is widely used in the treatment of hepatitis b patients in clinic. But if long-term application of it, hepatitis B patients may appear some side reactions, especially spiritual reactions. This can affects the patients'compliance and therapeutic effect, and limits clinical application of interferon alpha. Studies have shown that there is a close relationship between depression and the reduce of p11 in some specific regions of brain. So this study intends to investigate the relevance between the depression and the level of p11 mRNA in PBMC of hepatitis B patients, to make clear the molecular mechanism of the depression caused by interferon alpha.Methods1. Grouping:We selected 42 hepatitis B patients of a hospital, who were treated with IFN-a according to the patients' condition. Divided into two groups by the score results of the Hamilton depression rating scale (HAMD) and self-test depression scale (SDS). The depression group has 20 cases and the other group has 22 cases with no depression. Separately, chooseing 20 cases healthy subjects as control group.2. Extraction of peripheral blood RNA:Collected three group subjects' peripheral blood respectively, extracted peripheral blood mononuclear cells by lymphocyte separation medium, extracted mRNA, reversed transcription to get cDNA,detected the level of pll mRNA by real-time fluorescent quantitative PCR.3.Longitudinal observation of the level of pll mRNA of the hepatitis b patients who appear depression after treating of interferon alpha:according to the patients who have depression after medication, analyze the changeable of pll mRNA before treatment,4 weeks after treatment and 8 weeks after treatment.4.Compared three group subjects' p11 mRNA's changeable situation with before treatment,4 weeks after treatment and 8 weeks after treatment.5. Statistical Analysis:Use SPSS 19.0 statistical software for data processing and analysis, the results are expressed as mean+/-standard deviation (x+/-s) represent, using single factor analysis of variance, two groups are compared using t test, the difference was statistically significant (p< 0.05).Results1 After 4 weeks with IFN-a treatment, subjects were divided into the depression group, non-depression group and healthy control group according to the scale analysis.Relative expression of pll mRNA of three groups were respectively 0.82±0.10,0.96±0.13, 0.97±0.14. The relative expression of pll mRNA of depression group is lower than non-depression group and the control group (p< 0.01);There was no significant statistic difference between non-depression group and control group (p> 0.05).2 In view of the patients with depression after treatment performance, pll mRNA expression before and after 4 weeks treatment were 1.01+/-0.1,0.82+/-0.1,this has significant statistic difference (p< 0.01); But p11 mRNA expression were 1.00+/-0.2, 0.96+/-0.1 respectively before and after treatment in patients with non-depression after treatment, there was no significant statistic difference (p> 0.05);Conclusionthe level of p11 mRNA in peripheral blood of hepatitis B patients treated with interferon alpha is reduced when depression happened.
