BackgroundThe mechanism of lung cancer is unclear,so it is mainly due to inadequate for the prevention and treatment of lung cancer.Therefore,lung cancer become a serious threat to people's health.STAT3 is the key signaling protein of many growth factors and cytokines in the organism,and plays an important role in cell differentiation,function,immune balance and tumor development.MicroRNA21 is a small molecule RNA of non encoding,and it is involved in the growth,metabolism,development and disease of the body,and is related to the occurrence,development,invasion and metastasis of many kinds of tumors.In recent years,the research on the interaction between miRNAs and STAT family members is more and more,especially the interaction mechanism between miR-21 and STAT3 signaling pathway has been recognized by the majority of people.It has been confirmed that STAT3 can be directly activated by miR-21,And the regulation of abnormal display in many kinds of tumors.What is the significance of STAT3 MiR-21/ signal transduction pathway in the development of lung cancer? And what is the effect of T lymphocyte infiltration in the tumor environment? What is the significance of clinical treatment of lung adenocarcinoma?At present,there is no report.We detected the expressions of miR-21 and STAT3 in human lung adenocarcinoma tissue by real-time fluorescence quantitative PCR(q RT-PCR)method,and detected the infiltrations of T4 and T8 cells in lung adenocarcinoma tissues by immunohisto chemical.By analysising the relationship between them we investigate the miR-21/STAT3 signal transduction pathway abnormalities of lung adenocarcinoma occurred the development effects and the clinical significance,and provide new ideas for further clinical treatment and prevention of lung cancer.Objective Investigate the effect and significance in lung adenocarcinoma and the lymphocytes in the tumor environment of miR-21/STAT3,by detecting the expression of STAT3 and miR-21 in lung adenocarcinoma occurred in the process of the development of the dynamic expression and the infiltration of T4 and T8 cells in the tumor environment.Methods All samples were obtained from June 2013 to 2015 June,Department of Pathology of Liberation Army No.91 hospital and Jiaozuo People's Hospital of Henan Province,surgical resection of 46 cases of lung adenocarcinoma tissues and cancer adjacent tissues,including metastatic cancer 34 cases;12 cases of normal lung tissues as control.All the tissues were fresh tissues,and were kept in the refrigerator or liquid nitrogen in-80?,and diagnosed by Pathology diagnosis.The samples were not included in patients with recurrent lung adenocarcinoma,or lung adenocarcinoma patients who were treated with chemotherapy prior to surgery.Mir-21 and STAT3 gene expression were detected by q RT-PCR method,and the expression of T4 and T8 cells were detected by immunohistochemistry method.The experiment results were analyzed by t test and single factor analysis of variance by SPSS17.0.Results1.Routine pathological diagnosisIn 46 cases of lung adenocarcinoma,including 34 cases of lymph node metastasis,and12 cases of normal lung tissue were used as the control group.All tissues were diagnosed by pathological diagnosis.2.STAT-3 protein and its gene expression The expression of STAT3 mRNA in different tissues was detected by q RT-PCR,normal lung tissue 0.319±0.015,tumor adjacent tissue 0.631±0.020,adenocarcinoma tissue 1.046±0.042,metastatic cancer tissue1.235±0.015.There are differences between the normal lung tissue and the the adenocarcinoma tissue(P<0.01),and the differences among the tumor adjacent tissue,the adenocarcinoma tissue and the metastatic cancer tissue were statistically significant(P<0.01).The expression of STAT3 mRNA in lung adenocarcinoma was higher than that in normal lung tissue and Carcinoma adjacent tissue.3.MicroRNA21 expressionThe expression results of micro RNA21 by real-time PCR are as follows: normal lung tissue 0.0018±0.001,tumor adjacent tissue 0.017±0.005,adenocarcinoma tissue0.098±0.036,metastatic cancer tissue 0.126±0.134.Analysis of variance between groups F=17.016,P<0.05.There are differences between the normal lung tissue and the the adenocarcinoma tissue(P<0.01),and the differences between the adenocarcinoma tissue and the metastatic cancer tissue(P<0.05).Mi R-21 showed low expression in normal lung tissue,and high expression in lung adenocarcinoma.4.Expression of CD3+,CD4+ and CD8+ lymphocytes in tumor tissuesThe number of CD3+ cells was 76.12% in normal tissue,and there was no significant change compared with the normal group,and there was a significant decrease in the adenocarcinoma group.The number of CD4+ cell in the normal group was 45.17%,and the tumor adjacent tissue and the adenocarcinoma tissue were lower than the normal group,with significant difference(P<0.05).CD8+ in the normal group was 28.78%,and the adenocarcinoma group was higher than the normal group,with significant difference(P<0.05).Conclusions The occurrence and evolution of lung adenocarcinoma may be related to the increase of miR-21 and the disorder of immune function in the body.
