BackgroundThe main reason for the high morbidity and mortality of lung cancer is the mechanism has not yet clear,so the lung cancer as one of the main diseases which cause serious damage to people's health.In non-coding small RNA molecules,MicroRNA21(Mi R-21)as one of the main involved in the growth,metabolism,development and disease inside the body,and involved in a wide variety of tumor occurrence,development,invasion and metastasis.Signal transduction and transcriptional activation factor 3(STAT3)is mainly in many growth factors and cytokines of the body signaling proteins,which in the function of cell differentiation,cell,the immune balance and plays a very important role in tumorigenesis.In recent years about MiRNAs and STAT signal transduction between members of the family of the relationship between research more and more,especially in MiR-21 and STAT3 signaling pathways interact to get the recognition of most people.It has confirmed that STAT3 can directly regulate MiR-21,and in a wide variety of tumor showed abnormal regulation.So far no specific report showed that MiR-21 / STAT3 signal transduction pathway in lung cancer occurrence and development of significance.This topic using real-time fluorescent quantitative PCR(qRT-PCR)method,through the detection of human lung adenocarcinoma tissues MiR-21,the distribution of STAT3,explore MiR-21 / STAT3 signal transduction pathway abnormality impact on the occurrence and development of lung adenocarcinoma and clinical significance,provides new thinking for clinical prevention and treatment of lung adenocarcinoma further.ObjectiveBy detecting MiR-21 / STAT3 in lung adenocarcinoma dynamic distribution in the process of occurrence,development,discuss MiR-21 / STAT3 signal transduction pathway in lung adenocarcinoma in the process of impact.MethodsAll specimens from June 2013 to June 2015,the people's liberation army 371 hospital inspection department,xinxiang central hospital inspection department,surgical removal of the 18 cases of normal lung tissues,69 cases of lung adenocarcinoma tissues and its tissue adjacent to carcinoma(cancer tissue 2-5 cm),51 cases of metastatic carcinoma.From all the different types of organization as the fresh tissue samples,in-80 ? refrigerator or kept in liquid nitrogen.Normal lung tissue,lung adenocarcinoma tissues and its tissue adjacent to carcinoma were diagnosed by pathology(before surgery after chemotherapy of patients with lung adenocarcinoma and recurrence of lung adenocarcinoma patients with pathological specimens are excluded).Use qRT-polymerase chain reaction(PCR)to detect MiR-21 and the expression of STAT3 gene and immunoblot Western blot method to detect the p-the expression of STAT3 protein.Of the experimental results by SPSS17.0package t test and single factor variance analysis and related statistical analysis.Results1.Routine pathological diagnosis: 18 cases of normal lung tissue;69 cases of lung adenocarcinoma tissues(including high differentiation of 29 cases,18 cases of differentiation and low differentiation 22 cases)and its tissue adjacent to carcinoma 69 cases,including 51 cases of lymph node metastasis carcinoma.2.The expression of MiR-21 qRT MiR-21-polymerase chain reaction(PCR)test results: expressed by different organizations in the normal lung tissue was 0.0017 + /-0.001;Tissue adjacent to carcinoma was 0.018 + /-0.005;Adenocarcinoma tissue was0.099 + /-0.035;Metastatic carcinoma tissue was 0.127 + /-0.135.Within each group between the expression of genes have significant difference(P < 0.05),compared to normal lung tissues and lung adenocarcinoma tissue has the extremely significantdifference(P < 0.01).Micrornas-21 genes present minimum expression in normal lung tissue,the tissue adjacent to carcinoma was increased,present high expression in lung adenocarcinoma tissue,present the highest expression of metastatic carcinoma tissues.3.The expression of STAT3 gene qRT-polymerase chain reaction(PCR)to detect the STAT3 gene expression in different tissue results: normal lung tissue was 0.318 + /-0.015;Tissue adjacent to carcinoma was 0.629 + /-0.020;Lung adenocarcinoma tissue was 1.047 + /-0.041;Metastatic carcinoma tissue was 1.236 + /-0.016.Within each group between the expression of genes have significant difference(P < 0.05),compared to normal lung tissues and lung adenocarcinoma tissue has the extremely significant difference(P < 0.01).STAT3 gene present minimum expression in normal lung tissue,the tissue adjacent to carcinoma was increased,present high expression in lung adenocarcinoma tissue,present the highest expression of metastatic carcinoma tissues.4.The expression of P-STAT3 protein protein imprinting technology Western-p-the expression of STAT3 protein blot method test results: 1.00 + /-0.76 in normal lung tissue,lung adenocarcinoma was 2.00 + /-1.23 in the adjacent tissues,lung adenocarcinoma tissues was 3.80 + /-1.24,pulmonary metastatic carcinoma tissues was4.16 + /-0.95.Within each group between protein expression has significant difference(P< 0.05),compared to normal lung tissues and lung adenocarcinoma tissue has the extremely significant difference(P < 0.01).P-STAT3 proteins present minimum expression in normal lung tissue,the tissue adjacent to carcinoma was increased,present high expression in lung adenocarcinoma tissue,present the highest expression of metastatic carcinoma tissues.5.The correlation between MiR-21 and STAT3 gene and p-STAT3 protein By statistical analysis,in lung adenocarcinoma group,MiR-21 and STAT3 between present positive correlation(r =-0.910,P < 0.01);MiR-21 and p-between STAT3 protein also presents positive correlation(r =-0.953,p < 0.01).The consistency of the existence of correlation between STAT3 and p-STAT3,both a positive correlation(r = 0.710,p <0.710)Conclusions1.Mi R-21 and STAT3 and p-STAT3 proteins were highly expressed in lung adenocarcinoma,p-STAT3 proteins may occur in lung adenocarcinoma plays a major role.2.The abnormal expression of MiR-21,STAT3 may be related to the occurrence of lung adenocarcinoma is closely related to the evolution process.
