Rhizoma Panacis Majoris is the rhizomes of Panax japonicus C.A.Mey.var.major(Burk.)C.Y.Wu et K.M.Feng,belongs to Araliaceae Panax family.This article has mainly studied the extraction,purification,physical and chemical properties,structure,antioxidant and anti-complementary activity of polysaccharides from Rhizoma Panacis Majoris.Hope to lay the foundation for Rhizoma Panacis Majoris industrialized developmet and new drug discovery.The main contents as follows:1.Response surface method(RSM)was employed to optimize the extraction conditions of polysaccharides from Rhizoma Panacis Majoris.In order to obtain the optimal processing parameters,a three-variable Box-Behnken designs(BBD)were applied for experimental designs.RSM analysis indicated the good correspondence between experimental and predicted values,the optimal conditions for the yield of polysaccharides were as follows: the ultrasound time is 31.15 min,extraction temperature is 92.50 °C,and the ratio of water to raw material is 40 m L/g.The maximum value(13.87 ± 0.16%)of the yield of polysaccharides was obtained under these optimal conditions.2.Depurated polysaccharides from Rhizoma Panacis Majoris,then graded it by alcohol and obtained 30%,50%,60%,80%,four kinds of polysaccharides.A high performance size elusion chromatography coupled with multi-angle laser light scattering and refractive index(HPSEC-MALLS-RID)system was used to determine molecular weight(MW)and molecular weight distribution.PM1,PM2,PM3 polysaccharide sample consists of two parts,the molecular weight(MW)of macromolecule part decreased with increasing of the concentration of alcohol,Chromatogram of PM4 sample showed a single symmetrical peak,indicating a hig purity,good uniformity with molecular weight distribution width of 1.89,is the object of the follow-up study of physical and chemical properties and pharmacological activities.3.BCA method was used to determine the protein content of polysaccharide,the protein content in the sample was 0.14%.HPLC was established for determination mannose composition of polysaccharides from Rhizoma Panacis Majoris.FT-IR spectra demonstrated obvious characteristic peaks of polysaccharides,the absorptions at 1024,1080 and 1153 cm-1 indicated ?-pyranose of the glucose residue.4.The antioxidant activities of polysaccharides from Rhizoma Panacis Majoris were investigated including scavenging activity of hydrogen radicals,ABTS radicals,and free radicals of superoxide anion in vitro.Polysaccharides from Rhizoma Panacis Majoris have strong activities of ·OH and ABTS scavenging,and the effect of scavenging were more remarkable than that of superoxide anion scavenging rate.5.The anti complement activity of the classical pathway of the polysaccharide from Rhizoma Panacis Majoris was determined by the method of cell hemolysis.The anticomplement activity was concentration related,but the overall activity was not satisfaction.
