ObjectiveOn the basis of Preliminary experimental study,the changes of aortic endothelial cells JNK protein was observed by electro acupuncture in the treatment of the high fat diet rats,explore and improve the internal mechanism of electro acupuncture improve insulin resistance,but also provide the reference for the clinical treatment of insulin resistance related diseases.MethodsIn In the environment of SPF laboratory standard,selected 24 male SD rats were randomly divided into 3 groups,8 rats in each group,blank group,model group and EA group.The blank group of rats fed with ordinary feed,while the model group and the electroacupuncture group rats were fed with high-fat diet.After feeding for 12 weeks,plasma insulin(FINS)and fasting plasma glucose(FPG)were measured in the rats of the three groups,and the insulin resistance index(HOMA-IR)was evaluated by the homeostasis model.When the model has been completed,feeding the 3 groups of rats as before and rats in electroacupuncture group were given acupuncture Neiguan,Zusanli,Sanyinjiao and Shenshu.Zusanli and the Sanyinjiao were used electro(dense wave,50 Hz,rarefaction wave,2 Hz,10/min frequency modulation),the strength in rat hindlimb slight jitter for degrees,20 times/min,1/d,for 4w.At the end of the treatment,we should do those texts again and measure TC,TG,LDL-C and HDL-C.The expression of JNK protein and the phosphorylation will use Westernblot method to detect.ResultsAfter 12 weeks,the data of the two groups between the blank group and model group,including fasting blood glucose(FPG),fasting insulin(FINS)and HOMA-IR,adopting statistical analysis,the difference was significant(P<0.01);the data of the two groups between the blank group and E-A group,including fasting blood glucose(FPG),fasting insulin(FINS)and HOMA-IR,adopting statistical analysis,the difference was significant(P<0.01).After treatment,compared blank group and model group results;FPG,FINS,TG,TC,and LDL-C in model group increased significantly(P?0.01),while HDL-C decreased significantly(P?0.01).HOMA-IR also increased significantly in model group(P<0.01).In addition,the expression of JNK protein and its phosphorylation level in model group were significantly higher than that in blank group(P?0.01).Compared E-A group and model group results:the electro-acupuncture group rats FPG,FINS,TG,TC,and LDL-C were significantly decreased(P<0.01),while HDL-C increased significantly(P<0.01);The HOMA-IR in the E-A group decreased significantly(P<0.01).In addition,the expression and phosphorylation of JNK protein in the E-A group was significantly lower than that in the blank group(P<0.01).ConclusionVerify again,the rats fed with high fat diet can induce insulin resistance in rats,and establish the model of insulin resistance.The rats fed with high fat diet could successfully induce the insulin resistance in the experimental rats and establish the insulin resistance model.Electroacupuncture can reduce the content of FPG,FINS,TG,TC and LDL-C in rats,and improve the content of HDL-C,so as to improve the disorder of glucose metabolism and lipid metabolism in vivo;At the same time,the expression of JNK protein and its phosphorylation level were lowered,the insulin signal transduction pathway was restored,and then improve the state of the insulin resistance and increased the sensitivity of insulin resistance on insulin in rats.
