Objective:In this thesis,we focus on studying the effect of supplemented dangguishao-yaos for the p-ERK and let-7 gene expression in breast epithelial cells of mammary gland hyperplasia rats through the animal experiment,and the pathological observation of mammary gland tissue in rats was also investigated.This study will provide guidance for the cure of breast hyperplasia and breast cancer by regulating liver and spleen throgh investigating the effect of Danggui Shaoyao San on the transformation,differentiation and apoptosis of breast epithelial cells.Methods:We choose 72 female,healthy and not pregnant SD rats with weight 180-200 grams.The rats adapt to the environment after one week through free drinking water,natural light.If no abnormal performance,72 SD rats were randomly divided into blank control group contained 16 rats and test group contained 56 rats.In the blank control group,the rats in the other groups were modeled by estrogen and progesterone.One month later,the rats in the experimental group were randomly divided into model group 1,model 2 group,dairy addiction group,angelica root powder group.Each group has 14 rats.At the same time,rats of model group 1 were sacrificed,and the rats were sacrificed.The histological changes were observed under high magnification microscope to determine whether the pathological model of mammary gland hyperplasia was successful.After the success of the model,drug treatment,respectively.After a month of continuous gavage,the rats in each group were observed and all the rats were sacrificed and the mammary gland tissue samples were collected.The diameter and height of the nipples were measured.The expression of let-7 gene and p-ERK protein expression in mammary gland epithelial cells were determined by immunohistochemistry.The histopathological changes of mammary gland in experimental animals were compared and analyzed.Results:Model group,rupixiao group,nipple diameter supplemented dangguishaoyaosan group rats were greater than the height of the blank group,P<0.01.Rupixiao group and large group supple-mented dangguishaoyaosan nipple diameter,height less than the model groou,P<0.01,statistically significant.Rupixiao group rats and supplemented dangguishaoyaosan nipple diameter and height were not stat-istically significant,P>0.05;statistically significant;The p-ERK gene expression of each experimental group were higher than blank grou-p,P<0.01,statistically significant;the p-ERK gene expression of supplemented dangguishao-yaosan and rupixiao groups were lower than the model group,P<0.01,statistically significant;the p-ERK gene expression of supplemented dangguishaoyaosan was lower tha-nrupixiao group,P<0.01,statistically significant.The let-7 gene expression of each experimental group were lower than blank grou-p,P<0.01,statistically significant;the let-7 gene expression of supplemented dangguishao-yaosan and rupixiao groups were higher than the model group,P<0.01,statistically significant;the let-7 gene expression of supplemented dangguishaoyaosan was higher tha-nrupixiao group,P<0.01,statistically significant.Conclusions:Supplemented Dangguishaoyaosan can reduce the proliferation and enlargement of mammary gland in rats,improve the local mammary gland tissue edema,reduce the proliferation of mammary gland,promote the expression of let-7 gene and inhibit the expression of p-ERK protein in mammary gland hyperplasia epithelial cells Role,and thus inhibit cell proliferation,differentiation,promote apoptosis,improve the proliferation of breast tissue state.The mechanism may be Supplemented Dangguishaoyaosan with nourishing liver,dampness and spleen,the effect of promoting blood circulation,can adjust the body’s blood and yin and yang of the dynamic balance,leading to achieving the purpose of treatment and improving breast hyperplasia.
