Allergic rhinitis(AR)is a common inflammatory disease of the nasal mucosa that results from seasonal or perennial responses to allergen.It is an inflammatory disorder of nasal mucosa,which is characterized by nasal itching,sneezing,rhinorrhea and nasal congestion.Pharmacotherapy and immunotherapy,as well as avoiding the allergen are the common treatments for AR.The main treatment is pharmacotherapy including H1-anti-histamines,anti-inflammatory,leukotriene-receptor antagonists and intranasal corticosteroids.Though chemical therapy playes an important role on relieving symptoms of allergic rhinitis,the high recurrence rate of AR is still not satisfactory.Immunotherapy is the only one that can change the progression of AR,but it costs long time and lots of money.Therefore,it is the choice of AR therapy to find the traditional Chinese medicine that can control the recurrence rate and reduce costs.Gan Fu(GF)is extract from glycyrrhiza(GA)and radix aconiti carmichaeli(RAC)according to chinese medicine prescriptions by the clinical treatment of AR.In this research,we studied the pharmacodynamics of GF by allergic rhinitis rats in vivo model and the possible mechanism were explored.The results could provide experimental basis for traditional chinese medicine development.1.The effects on AR and cytokines in ratsThe AR model was induced by intraperitoneal injection of ovalbumin(OVA)in rats.Allergic symptoms were observed.Eosinophils(EOS)number of nasal mucosa with HE staining was counted.The serum concentration of IL-4,Ig E and INF-? was measured using ELISA method.(1)The results showed that OVA could induce allergic symptom such as scratching nose,runny nose,sneezing and obvious pathological changes in mucous membrane.OVA also could increase the number of EOS and the serum concentration of IL-4,Ig E and decrease INF-? concentration.(2)GF(0.25 g/kg and 0.5 g/kg)could significantly reduce the symptoms of rats with allergic rhinitis and reduce pathological damage of the nasal mucosa by gavage for 14 days.GF(0.5 g/kg)could significantly reduce the number of EOS.There was significantly difference compared with the model group.(3)GF(0.5 g/kg)could significantly increase the serum concentration of INF-y and reduce the serum concentration of IL-4 and Ig E.The results suggest GF has therapeutic action on allergic rhinitis.2.The anti-inflammatory action of GFWe observed the effects on ear swelling and capillary permeability of skin in mice induced by xylene.(1)The results showed that GF(0.5 g/kg and 1.0 g/kg)could significantly reduce the degree of ear swelling by gavage for 7 days compared with the model group.(2)GF(0.5 g/kg and 1.0 g/kg)could significantly reduce capillary permeability of skin in mice induced by xylene compared with model group.The results suggest GF have good anti-inflammatory effects.3.The effects of GF on the respiratory systemWe observed the effects on cough induced by ammonia in mice and phenol red secretion in bronchus of mice.We also observed the effects on Asthma of guinea pigs induced by acetylcholine chloride-phosphoric acid histamine.The GF groups were given a gavage of GF one time every day for 7 days and compared with model group.(1)The results showed that GF(0.25 g/kg and 0.5 g/kg)could significantly Prolong the latent period of cough in mice induced by ammonia;GF(0.25 g/kg,0.5 g/kg and 1.0 g/kg)could significantly reduce the frequency of cough in 3 minutes.(2)GF(1.0 g/kg)could significantly increase the amount of phenol red secretion in bronchus of mice.(3)GF(0.25 g/kg and 0.5 g/kg)could significantly Prolong the latent period of asthma induced by acetylcholine chloride-phosphoric acid histamine in guinea pigs and reduce the number of falls animals.The results suggest GF have significantly effects of relieving cough,eliminating phlegm and anti-asthma.4.The effects of GF on endurance in miceHypoxia mice model and swimming model in mice with weight loading were used to observe the effects of GF on endurance in mice.Cold resistant model in mice was used to observe the effects of GF on cold resistant ability of mice.The GF groups were given a gavage of GF one time every day for 7 days and compared with model group.(1)The results showed that GF(1.0 g/kg)could significantly prolong survival time in the condition of hypoxia.(2)GF(0.5 g/kg and 1.0 g/kg)could significantly prolong the time of swimming of mice with tail weight loading.(3)GF(0.5 g/kg and 1.0 g/kg)could significantly reduce the mortality rate in mice under the condition of five degrees below zero.The results suggest GF could significantly increase the endurance and resistance.LonclusionThe results suggest GF have therapeutic action on AR and anti-inflammatory eff-ect.GF have significantly effects of relieving cough,eliminating phlegm and anti-asthmatic.GF could significantly increase the endurance and resistance.
