ObjectiveThis experiment use scopolamine, sodium nitrite and ethanol respectively to set up mice model of acquired memory impairment,the consolidation of memory impairment in mice mode and reproduction-related memory impairment in mice model, with the experimental platform to observe Ditan Decoction on the impact of learning and memory function.MethodsSeventy-two Kunming mice, male, were randomly divided into three groups: the normal group (A),model group (B) and Ditan Decoction group (C). Each group received 0. 9%Nacl,0. 9%Nacl and Ditan Decoction once a day for three weeks. Use platform method to train and test learning and memory in each group. Detect apoptosis rate by flow cytometry in mouse brain cell, detect AchE expression by immunohistochemistry in the hippocampus, Detecte dopamine content by High Performance Liquid Chromatography in brain.Results1. Experimental platform show that:1. 1 Access to learning and memory impairmentModel group in the training and testing process, errors are more than normal group, the incubation period was significantly shorter than the normal group, and we find significant differences (P<0.01). Show that the memory access barrier to the production model is successful. Training and test results of Ditan Decoction group are significantly better than the model group (P<0. 01). Compared with the normal group, we find significant differences with training results of Ditan Decoction group (P<0. 05), and no differences with test results. It showed that Ditan Decoction has a significant improvement of mice model of acquired memory impairment.1. 2 The consolidation of learning and memory impairmentWhen the model was not built up, training results of Ditan Decoction group were significantly better than the model group and normal group (P<0. 05). there was no significant difference of the training results between normal group and model group (P>0. 05) .It showed that Ditan Decoction prevented memory impairment. Compared with normal group, the numbers of errors in test results of model and Ditan Decoction group were markedly increased,incubation period was short (P<0. 01).It showed that the model was successfully built. Test results of Ditan Decoction group were better than the model group, which showed that Ditan Decoction had Inhibition with the consolidation of learning and memory impairment which caused by sodium nitrite.1. 3 Representation of learning and memory impairmentwhen the model is not built, test results of Ditan Decoction group were better than the normal group and model group (P<0. 05). There were no differences between training results and test results of the normal group. However, training results were better than training results(P<0. 01).It showed that the model was successful. After the model was set up, test results of Ditan Decoction group were better than model group. There was no significant differences between the normal group and Ditan Decoction group (P<0. 01).It showed that the improvement of Detan Decoction to representation of learning and memory impairment.2. Detection of rate of neuronal apoptosis by flow cytometry methodRate of neuronal apoptosis of normal group was 1.189±0. 889%. rate of neuronal apoptosis of model group was significantly higher for 3. 986±1. 195%. Compared with the normal group, there were significantly different (P<0. 01). It showed that the memory impairment model was successful. Rate of neuronal apoptosis of Detan Decoction group was 1. 659±1.078%, significantly lower than model group(P<0. 01). Compared with the normal group, there was no significantly differences ( P>0. 05 ) . It showed that Ditan Decoction can significantly inhibited the role of neuronal apoptosis.3 . Impact on the expression of AchECompared with normal group, stained yellow of membrane and cytoplasm of hippocampal neurons of model group was significant deeper. It showed that expression of AchE of model group increased. Stained yellow of membrane and cytoplasm of hippocampal neurons of Ditan Decoction group was significant lighter than the model group and slight deeper than the normal group. It showed that expression of AchE is significantly decreased. Compared with normal group, Integral optical density of the model group increased. However compared with the normal group, Integral optical density of Ditan Decoction group decreased. There was no significant difference. It showed that Ditan Decoction can decrease the expression of AchE.4. Detection of the content of DA by High Performance Liquid Chromatography methodCompared with the normal group, the content of DA in mouse brain of the model group was significantly decreased (P<0.01 ) .However, Ditan Decoction can inhabit the decline of the content of DA. There were differences of the content of DA between the model group and the Ditan Decoction group.Conclusions1. Scopolamine,sodium nitrite and ethanol were respectively to set up mice model of memory impairment by blocking the three links of memory. The damages of memory were indeed, and the damages to taches were clear. Moreover, the method was simple. This model was a sophisticated and reliable model of memory impairment.2. Ditan Decoction could significantly improve the memory acquisition and memory retrieval in mice .However, improvement of memory consolidation is slight.3. Ditan Decoction has a role in prevention and improvement of memory and learning by increasing the content of DA of brain tissue, reducing the expression of AchE and suppresses apoptosis of neurons.4. "Sputum" theory of Traditional Chinese Medicine is associated with the occurrence and development of memory. The methods of transforming phlegm and opening the orifices are the basic treatment which could prevent and improve memory impairment. |
