Objective Edema is the main symptom of Nephrotic syndrome(NS),and the lung and spleen and kidney are closely related to edema.Combined with traditional Chinese medicine theory and modern medicine on aquaporin(aquaporin,AQP)understanding,that the qi and spleen water method to treat NS and aquaporin regulation of the correlation.Studies have shown that the representative side of Shenlingbaizhu powder on the kidney aquaporin has a strong regulatory role.The effects of Shenlingbaizhu Powder on the regulation of aquaporin and the effects of Shenlingbaizhu powder on the rat model were investigated by observing the expression of aquaporin in rat model of adriamycin nephrotic syndrome.The Methods 50 SD rats were divided into 5 groups: blank control group,model group,Shenlingbaoshao low dose group,Shenlingbaizhi powder middle dose group and Shenlingbaizhi high dose group.Among them,the model group,Shenlingbaizhu powder low dose group,Shenlingbaizhu powder in the middle dose group,Shenlingbaizhu high dose group were used spleen qi deficiency type adriamycin nephropathy model.The rats were treated with Shenlingbaizhu San as carriers.After the end of the experiment,serum biochemical markers and urine protein were measured.And AQP1/2/3/4 expression in renal tissue was detected by immunohistochemistry.The expression of AQP1/2/3/4 in renal tissue was detected by immunoblotting and the mRNA expression of AQP1/2/3/4 was detected by real time-PCR the amount.Results 1.Compared with the model group,the serum levels of total cholesterol(TC),triglyceride(TG),triglyceride(TG),triglyceride(TG),triglyceride(TG),triglyceride(Creatinine,CRE),blood urea nitrogen(BUN)and sodium,chloride and potassium ions were used to regulate the total protein(TP)and albumin(ALB).2.The expression of AQP1/2/3/4 in the kidney tissue of rats with Spleen Deficiency type adriamycin nephropathy was down-regulated by Shenjiapingji high dose group.3.Immunofluorescence assay showed that overexpression of AQP1/2/3/4 in thekidney tissue of rats with Spleen deficiency type adriamycin nephropathy was down-regulated in the high dose group.4.At the gene level,the Real Time PCR method showed that the expression of AQP1/2/3/4 mRNA in the kidney tissue of rats with Spleen deficiency type adriamycin nephropathy was down-regulated in the high dose groupConclusion The expression of AQP1/2/3/4 in renal tissue was significantly higher than that in NS group Angle treatment of Shenlingbaizhu powder treatment of NS,to improve the edema provides a theoretical basis. |
