Objective: To investigate the effect of white hyacinth bean polysaccharide on liver morphology,serum and biochemical indexes in alcoholic liver disease rats,and explore the possible mechanism of it.Methods: 90 male SD rats were randomly divided into normal group,model group,positive control group of metadoxine,with low dose group,middle dose group,high dose group of white hyacinth bean polysaccharide?n=15?.Method of creating the model of alcoholic liver disease is that feeding rats with high count liquor and high lipid diet,except for normal group.Two hours before the rats in white hyacinth bean polysaccharide groups and mtdx group gavaging liquor,the rats were treated with white hyacinth bean polysaccharide 0.5 g·kg-1,1.0 g·kg-1,2.0g·kg-1and metadoxin 0.09 g·kg-1.The normal group received normal diet every day,while the others fed high fat diet.After 10 weeks,all the rats were killed and the blood and liver were taken.1.Following indicators were detected to investigate the preventive and therapeutic effects of white hyacinth bean polysaccharides on ALD rats.The liver index of the rats was measured by weighting the wet liver.Pathological changes of liver tissue were observed by HE staining.The contents of AST and ALT in serum were measured by microplate method.The contents of TG and TC were detected by using enzyme standard colorimetric method.2.Following indicators were detected to investigate the antioxidative mechanism of white hyacinth bean polysaccharide on ALD rats.The activity of SOD in liver tissue was detected by xanthine oxidase method.The activity of GSH-Px was detected by colorimetric method.MDA content was detected by thiobarbituric acid method.Western blot was used to detect the activity of CYP2E1 The expression level of protein and mRNA relative expression.Result:1.Compared with normal group?2.020±0.0987?,the liver index?2.731±0.0681?was higher in the model group?aP<0.05?.The liver index of the white hyacinth beanpolysaccharide groups were?2.431±0.0927?,?2.436±0.188?,?2.206±0.2039?.Compared with the model group,the liver index was decreased in each dose of white hyacinth bean polysaccharide groups?bP<0.05?.Compared with metadoxine group,the liver index of rats in white hyacinth bean polysaccharide group decreased?cP<0.05?.2.The results of HE staining: the normal morphology of liver cells are clear boundary,arranged in a radial arrangement along the manifold area;the border of rat hepatocytes were blurred,the cytoplasm containing many fat vacuoles with varying sizes in model group,and different degrees of periportal lymphocytic infiltration were seen in the portal area;compared with the model group,the number of fat vacuoles in liver cells in white hyacinth bean polysaccharide groups were significantly reduced.And the morphology of cells was improved,especially in the high dose group,the morphology of cells was closest with it in normal group.The cell morphology of metadoxine group was similar to the model group.3.The content of ALT level in serum was?45.25±4.652?U/L in the normal group and?97.13±14.211?U/L in the model group,which was significantly higher than that in the normal group?aP<0.05?.The low,medium and high doses of white hyacinth bean polysaccharide group were?66.63±7.050?U/L,?70.00±11.36?U/L,?64.25±7.451?U/L,which were significantly lower than those in the model group?bP<0.05?U/L.The metadoxine group was?72.75±7.517?U/L,was no significant difference with white hyacinth bean polysaccharide groups?c P>0.05?.4.The content of AST level in serum was?125.4±7.94?U/L in the normal group and?211.6±17.18?U/L in the modle group,which was significantly higher than that in the normal group?a P<0.05?.The low,middle and high dose groups of white hyacinth bean high dose of white hyacinth bean polysaccharide groups?cP<0.05?.Conclusion:1.we gave spirit by gavage and fed high fat diet to rats for making the model of alcoholic liver injury in this experiment,which could make the Clinical symptoms of ALD appear in rats.2.The white hyacinth bean polysaccharide could reduce the liver index and the levels of ALT,AST,TG and TC in serum of ALD rats,and improve the histopathological morphology of liver tissue,which showed that it had certain protective effects on liver tissues damaged by ALD.3.The white hyacinth bean polysaccharide can not only increase the expression of GSH-Px and SOD,but also reduce the expression of MDA,and inhibit the expressionof CYP2E1 protein and gene,which told us that the white hyacinth bean polysaccharide can prevent the injury of hepatic tissue in ALD,the possible mechanism is that the white hyacinth bean polysaccharide can anti lipid peroxidation injury through microsomal ethanol pathway.
