ObjectivesAir pollution has become a hot topic of increasing concern,its extensive sources of pollution,diffuse into every corner with the meteorological factors.The surface of the pathogenic microorganisms and transition metal ions can lead to respiratory tract bacterial flora disorder,leading to micro-ecological disorders,causing some endogenous infection.In this study,we investigated the effect of different doses of atmospheric mixed pollutants on the SHR rat model,and the effects of different doses of atmospheric mixed pollutants on the respiratory microflora and the changes of respiratory tract flora.Detection of serum related cytokines in SHR rats.Determination of key gene expression levels of VEGF metabolic pathway in cardiovascular system.The aim of this study is to provide the basis for the damage caused by fine particles to the body.In order to lay the foundation for the study of microbial population,immune function and mechanism of cardiovascular system damage to the body.Methods?According to the national environmental quality standard two times,10 times,100 times,The animal models were made by challenging SHR/NCrl rat by the current status of PM2.5 exposure,?The systolic blood pressure of the tail artery was measured with a noninvasive blood pressure instrument(BP2010A).? The levels of[Interleukin-6(IL-6)],[Interferon-?(IFN-?)],[tumor necrosis factor(TNF-?)]in the serum of SHR/NCrl rats were measured by ELISA.At different time(Id,7d,30d,2M,3M).?Microbial culture and identification of microflora in oropharynx of SHR/NCrl rats at different time-point poisoning(1d,7d,30d,2M,3M)?The expression of Hsp27,PLCy and VEGF in the metabolic pathway of cardiovascular system in SHR/NCrl rats were measured by by fluorescence real-time quantitative PCR based on the transcription differentially expressed genes.Results?Dust exposure dose increased the blood pressure rise,may cause vascular endothelial dysfunction.?comparison of different cytokines in rat serum,The results of ELISA showed that the expression of TNF-?(P<0.01)was significant difference between the different dose groups and the control group in the serum of the rats.The difference was significant at 3M(P<0.001).There was significant higher between the 1L-6(P<0.05)high dose group and the control group.The difference was significant at 3M(P<0.001).IFN-? was significantly different from the control group at 1d(P<0.001).?The detection rate of anaerobic bacteria in SHR rats was 71.8%and 20.7%respectively,compared with the previous exposure to dust was high than 0.6%,there were significant differences(P<0.05).In the detection rate of anaerobic bacteria,the Tetanus bacillus and Clostridium in the respiratory tract of the exposed to dust rats was not detected.After the dust exposure,two kinds of bacteria respectively in 28 and 5 SHR rats were detected.The detection rate of Veillonella in anaerobic bacteria was 53.1%,there were significant differences(P<0.05).The number of pathogenic bacteria in the aerobic bacteria after the dust exposure was increased,the detection rate also increased.The results showed that in the body the number and species of bacteria after the exposure to dust were changed.The results showed that,After the dust treatment,the aerobic bacteria(P<0.05)and the total bacteria(P<0.05)showed significantly different.The detection rate of the pathogen(P<0.05)was increased.?The expression of Hsp27,PLCy and VEGF in different doses of SHR/NCrl rats was significantly different(P<0.05).The relative expression of VEGF gene was increased with the time period,and the relative expression of VEGF gene in middle and high dose group was about 2 times lower than that of low dose group.The relative expression of PLCy and Hsp27 increased and decreased.Conclusion?In this experiment,the model of Spontaneously Hypertensive Rats was established by scientific sampling,preparation of experimental materials and samples.In the mixed dust was exposed to atmospheric pollutants,elevated blood pressure in rats,endothelial function disorder.?The levels of IL-6 and TNF-a in the serum of the experimental group and the control group were detected by ELISA.The content of IL-6 and TNF-a in serum could be used as one of the bio-markers of the inflammatory reaction before the organism.Indicating that the body appears with the immune injury of inflammation and oxidative stress.?The relative expression of Hsp27,VEGF and PLC? in VEGF metabolic pathway can be used as one of the bio-markers of cardiovascular system injury.Confirmed that changes in serum bio-markers can be used as an early indicator of changes in the expression of cardiovascular system-related genes,may begin in the cascade of inflammatory response.?bacterial infection may be caused by cardiovascular disease in viral myocarditis and atherosclerosis reasons.Air pollution may be a complex process of injury on the cardiovascular system,the adverse reactions are by a variety the superposition of factors and ways caused.?PM2.5 as the carrier of harmful substance,when with breathing enter the human respiratory tract,will cause the normal flora in the body's respiratory tract disorder.The pathogen bacteria is easy to be colonized and the detection rate is increased,which affecting the body healthy.Inflammatory response factors as an intercellular signal,transmission and mediated chronic micro inflammation.When the micro environment of the body is broken,the balance between the microbes,the host and the environment is broken,breaking the physical,biochemical and immune barrier of the oropharynx of the body,and the oropharynx and the respiratory tract are connected so that the respiratory system is infected.Respiratory microbial balance is broken,the density of bacteria changes,pathogens easy to colonization,At the same time,the respiratory micro-ecological balance of many lines were broken,so that the original bacteria group disorders,leading to micro-ecological disorders.
