Sleep deprivation can provide a basis for understanding the mechanism of sleep disorders as an example of extreme sleep reduction.Default mode network(DMN)is posited to play a fundamental role in maintaining the resting state functional brain activity and involves a variety of cognitive process such as sleep and neuropsychological diseases.But whether the effect of selective REM sleep can deprive the effect on the DMN network is unclear.In this study,synchronized local field potential were recorded and rats were subjected to 4 hours REM sleep deprivation,which was based on the low-pressure REM sleep deprivation method,to explore the changes of sleep stage,EEG power spectra,DMN network and network attributes,in order to further understand the neurological mechanism of DMN network after sleep deprivation.The main results are as follows:1.Through the sleep stage analysis,we found that REM sleep could rebound after sleep deprivation,mainly for the significant increase of REM sleep time and number,was a compensatory response of REM sleep.2.Through the whole brain power spectrum analysis,we found that,after sleep deprivation,REM power spectrum changes were most significant in the gamma band,with a significant increase.And NREM power spectrum changes in the delta,theta,alpha,beta,gamma band were most significant,with significant reducing.The power spectrum of different brain regions was analyzed and we found it was consistent with the whole brain power spectrum.3.Through the DMN analysis,we found that,(1)after sleep deprivation,the delta's network connection of REM taken weakness,of which prelimbic cortex was the most significant.And DMN connection was improved in the theta band's parietal subsystem,of which hippocampus and temporal lobe were the most significant.Through the analysis of network attributes,delta band clustering coefficient was decreased,the characteristic path length was increased,and the small world attribute was decreased.But the theta band showed the contrary change.Overall,the trend of delta and theta band appeared gradually recover.(2)Sleep deprivation had delay effect on NREM's delta and theta band.And DMN connection were improved in the delta and theta band,of which prelimbic cortex and orbital frontal cortex were most significant.Through the analysis of network attributes,delta band clustering coefficient was significant increased,the characteristic path length was significant decreased,and the small world attribute was significant increased.But there is no obvious change in the theta band.Overall,the trend of delta band appeared gradually recover.The change of REM and NREM's DMN network may reflect that sleep deprivation had different effect on the information transmission rate of DMN network of REM and NREM.This study based on rat REM sleep deprivation method,illustrated the effect of REM sleep deprivation on the DMN network and network attributes of REM and NREM.Our results support the viewpoint that sleep play an import role on the brain functional networks.We also provided a new theoretical basis to understand the network mechanism of sleep,and helped to futher understand the neural mechanisms underlying sleep disorder.
