Viral respiratory tract infection is a major component of respiratory infections.The interferon induced transmembrane family is involved in a wide range of cellular events.There are 5 kinds of human IFITMs family proteins,IFITM1,IFITM2,IFITM3,IFITM5 and IFITM10,all located on chromosome 11.The IFITM family can be divided into three groups,one is immune related IFITM1,IFITM2 and IFITM3,which are involved in the cellular immune phase of resistance to viral invasion.The second is bone restriction protein IFITM5,which is involved in the deposition of calcium salts in bone.The third type of IFITM10 may be related to water environment adaptability.In the mouse IFITMs family,there were IFITM6 and IFITM7,except for IFITM7 in chromosome 16,and the rest on chromosome 7.The IFITM family regulates cell proliferation and promotes cell adhesion.They are overexpressed in a variety of tumors and are closely related to tumor cell migration,invasion,and proliferation.IFITM1,IFITM2,and IFITM3 are involved in the invasion of viruses.IFITMs localized on the surface of the cell membrane.IFITM1 is most effective against filamentous viruses,and IFITM3 is most effective against influenza A.IFITM3 were located in the late endosomal pathway dependent on IFITM3 protein in the membrane region 1 mediated activity,and through the end of N-and IFITM protein in the conservative intracellular loop together,the maximum limit of virus invasion.The IFITMs family plays an important role in life activities,and the functional roles and mechanisms involved in the organism need to be further studied.Objective:To analyze the difference of IFITMs expression in the samples of respiratory infection,and to explore the relationship between IFITMs protein and virus.Objective to study the expression of IFITMs gene in rat tissues.Methods:1.Summary of Clinical information classification and respiratory virus infection werecarried out on the samples of lower respiratory tract infection.2.Human IFITMs primers were designed and screened for amplification of IFITMs byRT-qPCR a in the lower respiratory tract infection samples,and the relativeexpression of IFITMs was analyzed.3.RNA was extracted from rats and IFITMs primers were designed and screened.Theexpression of IFITMs in 32 tissues of rats was analyzed by RT-qPCR.Results:In this study,320 children with lower respiratory tract infection were analyzed.There was a statistically significant difference in the expression of IFITM3 between bronchopneumonia and severe pneumonia,mild pneumonia,and severe pneumonia.The ability of IFITM3 to control influenza viruses and the restriction ability of IFITM1 against coronavirus are different from other viruses.Nucleic acids extracted from bronchoalveolar lavage fluid from patients with respiratory infection were found to have an expression of IFITM5 at the mRNA level.The expression of IFITMs and mRNA in 32 tissues of rats showed that the expressions of IFITM1,IFITM2 and IFITM3 were more common in all groups.In the IFITM5 expression map,it was found to be expressed in tissues other than the bones,such as bone marrow,skin,pancreas,and smooth muscle.The relative expression of IFITM6 in eyes is the highest,and IFITM2 and IFITM7 have similar expression trends.The expression of IFITM3,IFITM5 and IFITM10 in smooth muscle was significantly higher than that in skeletal muscle.Conclusions:The relative expression of IFITM3 protein changed during exacerbation of respiratory tract infection.Different IFITMs members differ in their ability to restrict the same virus.The mechanism of IFITMs restriction on virus invasion needs further study.In the IFITMs expression lineage of rats,there were differences in the expression of IFITM5 in bone.Because of homology,IFITM2 and IFITM7,IFITM5 and IFITM10 have many similarities in the tissue expression map,but their function in the tissue needs further study.
