Influence Of Ganweibaihe Decoction On Intervention In Th1/Th2 Cells Balance Of Chronic Stress Gastric Ulcer Mice Mediated By JAK/STAT Pathway

Objective:Discuss the influence of Ganweibaihe decoction on Th1/Th2 cells balance of chronic stress gastric ulcer(liver-stomach and qi stagnation syndrome)of mice.Clarify the regulation Ganweibaihe decoction on endocrine,immune system and repair-protection mechanism on the gastric mucosa damage.Methods:Randomly divided 40 mice into four groups:blank group,model group,western medicine group and traditional Chinese medicine(TCM)group,each10 included.Use 10 kinds of unpredictable stimulation factors manufacturer chronic stress gastric ulcer mice model,on the 14 th day,the blank group,model group were given distilled water,the western medicine group,TCM group were given corresponding drugs.Until 28 th days collect sample,scored open-field-test results,caculated ulcer index,adopted ELISA to test the level of inflammatory factors and immunohistochemical to test JAK2/STAT4 protein expression.Results:1.General situation: after modeling,model group,TCM group and Western medicine group were poor mental state,lack of appetite,hair gloss,water consumption decreased significantly,like aggregation,slow response,dark stool,urine slightly yellow.After given the corresponding drugs,the model group had no obvious improvement as the TCM group,western medicine group has improvement.2.Open-field-test:Molding 14 days,compared with the blank group,the horizontal score,vertical score of model group,western medicine group,TCM group are dramatic decline,the-central-lattice-residence-time shortened,with significant difference(P< 0.01).After 14 days of treatment,compared with model group,TCM group and Western medicine group were increased with statistically significant(P <0.01);the TCM group was higher than the western medicine group,the difference was statistically significant(P < 0.05).3.Ulcer Index:To observe the TCM group and western medicine group of mice gastric mucosa HE staining,only a small amount of erosion,erosion and scattered in the model group showed punctate ulcer;Compared with model group,TCM group and western medicine group have significantly lower UI,with significant difference(P<0.01).Compared with western medicine group,TCM group has lower UI,difference was statistically significant(P<0.05).Ulcer inhibition rate of TCM group is obvious higher than western medicine group.4.Compared with the blank group,the level of IL-2,IFN-?of model group are significantly increased,the level of IL-4,IL-10 are significantly decreased,the difference was statistically significant(P<0.01).Compared with model group,the content of IL-2,IFN-? in serum of TCM group and western medicine group are reduced,the content of IL-4,IL-10 are rise,with statistically difference(P<0.05).TCM group and western medicine group,the difference has statistical significance(P<0.05).5.Compared with the blank group,the levels of JAK2 and STAT4 in model group were obviously increased,the difference was statistically significant(P<0.01).Compared with the model group,the level of JAK2 and STAT4 in the western medicine group and TCM group decreased obviously,the difference was statistically significant(P < 0.01);the level of JAK2 and STAT4 in the TCM group was lower than that of the western medicine group,the difference was statistically significant(P < 0.05).Conclusion:1.Successfully cloned the chronic stress gastric ulcer mouse model.2.Under chronic stress stimulation,endocrine-immune system of mouse was stimulatd,released more inflammatory factors,changed the internal homeostasis of normal body and induce Th1/Th2 cells out of balance,stimulate the immune response.Suggests that chronic stress can change the gastric mucous membrane barrier function and immune response,damage the gastric mucosa,generate gastric ulcer.3.The mechanism of Ganweibaihe decoction treat chronic stress gastric ulcer,may through reduce the JAK/STAT expression,reduce the production of IL-2,IFN-?,on the other hand,increase the production of IL-4,IL-10,regulating the balance between Th1/Th2 cells,prompted the immune response normally process,alleviate immune response and the injury of inflammatory factors to gastric mucosa,promote the repair of the gastric mucosa and ulcer healing.
