Objective To investigate the expression of micro RNA-638(mi R-638)in malignant gliomas tissues/cells,and to observe the effect of mi R-638 expression on the proliferation,migration and invasion of U251 cells.Preliminary detect mi R-638 Inhibits Invasion of Glioma Cells via Targeting Notch 4.Methods(1)RT-PCR was used to detect expression of mi R-638 in normal nerve tissues/cells and glioma tissue/cells.(2)U251 cells(mi R-638 expression significantly)were used to transfect of mi R-638 mimics and inhibitors of mi R-638,the invasion level was detected by Transwell assay;(3)Cell proliferation and migration ability was assessed by MTT and cell scratch test respectively after transfection of mi R-638.(4)Targeting relationship between mi R-638 and Notch4 was predicted and validated by bioinformatics analysis.(5)Overexpressing Notch4 by constructing eukaryotic expression plasmid and transfected into U251(p CMV-Notch4 c DNA),invasion level of U251 cells was detected by Transwell assay.Results(1)Expression of mi R-638 in glioma tissues and cell lines was up regulated;(2)Invasive ability of U251 cells can be promoted by mi R-638 mimics,Vice effect instead(P<0.01);(3)Mi R-638 promoted the migration of U251 cells,but did not affect U251 cells proliferation capacity(P > 0.05).(4)Notch 4 was down-regulated in U251 cell,the correlation was negative with mi R-638;(5)In U251 cells,mi R-638 target to inhibit the expression of Notch 4 directly,enhance the expression of Notch 4 in U251 cells also can reduce the number of invasive cells(P<0.01).Conclusion(1)Mi R-638 is highly expressed in gliomas,which may promote the invasion and migration of glioma cells.(2)Mi R-638 may promote the invasion of glioma cells by inhibiting the expression of Notch 4.
