Abstract/Summary:
Objective:To study the regulation effect of ginseng mume Decoction on urine metabolism of diarrhea model rats by 1H-NMR metabolomics methods,and to explore the mechanism of ginseng mume Decoction’s function of replenishing qi and generating fluid anti-diarrhea effect,by the way, it provide in-depth theoretical reference and experimental basis for further revealing Chinese herbal compound of complex systems with multiple levels and multiple targets and a comprehensive understanding of the efficacy of traditional Chinese medicine prescriptions.Experimental method:Select 48 SD young rats,12 were randomly divided into blank group, model group 36, making complex model of diarrhea by the bitter cold diarrhea with weary. After successful model production,the rats were randomly divided into model group, western medicine group, TCM group,12 in each group,then started administration intervention. The blank group and model group were given normal saline 30mL/kg to fill the stomach, Other groups were given intragastric administration of 35g/kg ginseng mume Decoction, MAMIAI 0.7g/kg,all 1 times Id, continuous administration for 7d.1) After treatment, rats were carried on general observation, stool detection, diarrhea rating and observed tissue morphological with light microscope to assess the effect of ginseng mume decoction on treating diarrhea.2)using 1H-NMR metabolomics methods to study the changes of metabolic profile and key metabolites in urine of rats after treatment and give a preliminary discussion on the curative mechanism of traditional Chinese medicine to improve the intestinal micro ecology through the host metabolic phenotype.Experimental results:1)General changes:After modeling, stool frequency in the model group was significantly increased, anal contamination, fur loss of smooth, slow growth in weight, With mental fatigue, irritability or restlessness is not easy, with mental fatigue, irritability or restlessness,lips, nails showed dry red, hair haggard, part prolapse occured in rats, and even blood in the stool. Each dose group of rats after dosing in mental state, activity, body weight,and diarrhea condition was lighter than model group, the signs and symptoms after medical treatment gradually reduced and disappeared.2)Histomorphology result:blank group:normal colon tissue structure, intestinal epithelium integrity, there was no obvious inflammatory cell infiltration, edema and injury.Model group:intestinal mucosal epithelium showed multiple necrosis, ulcer, mainly affected the epithelial layer, the gland has no obvious damage; obvious congestion, edema and infiltration of inflammatory cells were seen in the submucosal layer, by the way,muscular layer has no obvious damage; TCM group and Western medicine group showed different degrees of improvement compared with the model group.3)Study on metabolic phenotype of rat urine in model rats:Multivariate statistical methods showed blank group, model group, western medicine group, TCM group was significantly separated,which the urine samples of traditional Chinese medicine group model was closer to the blank group than the model group. The factor loading chart was used to analyze the cause of separation effect of the metabolites with the blank group, TCM group of deviation of aconitic acid in the urine, creatinine, leucine and isoleucine, alanine, citric acid, acetone, has obvious positive adjustment.Conclusion:Ginseng mume Decoction to the general behavior of diarrhea rats have obvious improvement effect and obvious therapeutic effection on diarrhea rats colonic mucosa ulcer and inflammatory invasion, for diarrhea models also have protection and restoration of intestinal mucosa.Ginseng mume Decoction have a positive moderating effect on the key metabolites in urine of rats, which aconitic acid, creatinine, leucine isoleucine, alanine, citric acid, acetone as the key metabolite of diarrhea.
