Investigation Anti-inflammatory Properties Of Fu Fu Brick Tea For Dextran Sulfatesodium-induced Ulcerative Colitis In C57BL/6 Mice

Ulcerative colitis (UC) is a kind of inflammatory bowel disease, its pathogenesis is not clear. In recent years, UC incidences in Eastern Europe, South America, Asia and other low-incidence areas have significantly increased. Fu Brick Tea is the special local dark tea products in Hunan province, it can enhance the intestinal immune function but only reported in epidemiological surveys. This study used oral administration different concentrations water extracts of Fu Brick Tea as treatments for DSS-induced acute UC in C57BL/6 mice, observed theirs relative weight variations, blood in the stools, food intakes, visceral indexes, colon tissue sections morphological indicators for C57BL/6 mice, detected the difference variation of serum inflammatory cytokines of TNF-a, IFN-y, IL-6, IL-8, IL-1?, measured the variations of relative expression levels of Stat3, Jak2, Bax, Bcl-2 mRNA gene in Jak2/Stat3 inflammatory pathways in colon tissue of C57BL/6 mice. Experimental results showed that:(1) Oral administration water extractions of Fu Brick Tea can improve living conditions of DSS-induced C57BL/6 UC model mice. Treatment with water extractions of Fu Brick Tea and EGCG altered their physics and mentality states to normal mice levels, recovered to normal dietary habits, normal defecate, and weight increasing steadily.(2) Oral administration water extractions of Fu Brick Tea to C57BL/6 UC mice made significantly impact for viscera relative weight and colon length. Compared with normal control group, the heart, liver, spleen relative weight were increased, the length of the colon was shorten, the relative weight of the kidneys was substantially no effected in C57BL/6 model UC mice; Oral administration different concentrations water extractions of Fu Brick Tea and EGCG, the heart, liver, spleen relative weight and the colon length has recovered to significant levels in C57BL/6 model UC mice.(3) Oral administration water extractions of Fu Brick Tea to C57BL/6 model UC mice made significantly pathological damage repair. Oral administration different concentrations water extractions of Fu Brick Tea and EGCG, colon tissue had a good recovery, colon epithelium became integrated, a small amount of epithelium fell off, the number of goblet cells were close to the normal group levels, intestinal glands were arranged neatly on the top of the lamina muscularis.(4) Oral administration water extractions of Fu Brick Tea played significantly roles to lightening the C57BL/6 model UC group mice serum inflammatory factors. Compared with normal control group, the expression of inflammatory factors for TNF-??IFN-??IL-6?IL-8? IL-1?in C57BL/6 model UC group mice serum were significant increased. These results explained that oral administration water extractions of Fu Brick Tea and EGCG have good inhibited pro-inflammatory cytokines for TNF-??IFN-??IL-6?IL-8?IL-1?.(5) Oral administration water extractions of Fu Brick Tea made significantly regulation on JAK2/STAT3 inflammatory pathways for C57BL/6 model UC group mice in colon tissue. Compared with normal control group, the expression of JAK2, Bax 2 gene mRNA levels in colon tissue of C57BL/6 model UC group mice were significant decreased, and STAT3, Bcl-2 gene mRNA levels were significant increased (P<0.01); Oral administration different concentrations water extractions of Fu Brick Tea and EGCG, the expression of JAK2, Bax 2 mRNA levels in colon tissue of C57BL/6 model UC group mice were significant increased(P<0.01), and the STAT3, Bcl-2 gene mRNA levels were significant decreased(P<0.05). DSS induced acute UC in mice leads to the ratio of Bax:Bcl-2 decrease in the colonic tissues of mice, oral administration different concentrations water extractions of Fu Brick Tea and EGCG, compared with the (1) normal group, the ratio of Bax:Bcl-2 increase with (5)?(8)group, the ratio of Bax:Bcl-2 decrease with (6) group, and the ratio of Bax:Bcl-2 with (7) group is close to the normal group.So, it could be concluded that DSS-induced C57BL/6 mice acute ulcerative colitis and oral administration water extractions of Fu Brick Tea can improve morphological indicators of the UC model mice body weight, food intake, blood stool situation, viscera index, colon tissue sections forms; make inflammatory factors of IL-6?IL-8?OL-1??TNF-??IFN-?levels recover to the normal control group; regulate the Jak2/Stat3 inflammatory metabolism pathway for mRNA JAK2, STAT3, Bcl-2 and Bax expression levels to the normal control group. Oral administration water extractions of Fu Brick Tea make obviously repair effects for DSS-induced C57BL/6 mice acute ulcerative colitis.
