Aim:Establish the rat model of radiation pneumonitis,take sang xing qingying decoction as the prescription to observe the pathological side effects of lung tissue, the level of serum proinflammatory cytokines IL-6,TNF-a;the lungtissue SOD activity and MDA, MMP-2,TIMP-2 content; the expression of TGF?1.Then assess its prevention and control effect of radiation pneumonia,and itspossible mechanism.Methods:The 180 male SD rats weight about 280g to 300g of clean grad e were randomly divided into normal control group,merely irradiated group,sang xing qing ying decoction prevention group.sang xing qing ying decoction treat ment group,western medicine group,each group of 45.In addition to the normal control group,other groups were given chest X-ray single-wide exposure 20Gy. Apart from sang xing qing ying decoction prevention group started 4 days bef ore radiation fed the decoction, the rest of the group the next day after model ing,the normal control group,model group were given the same volume of nor mal saline,sang xing qing ying decoction treatment group fed sang xing qing y ing decoction, western medicine group were given prednisone once a day.Each group sacrificed 9 rats at 2,4,8,12weeks,took the blood after anesthetized rats; c ollected lung tissue of rats, calculating lung coefficient;HE stained pathological changes of lung tissue;observed by ELISA assay of serum IL-6, TNF-a content; lung tissue MDA,SOD,MMP-2,TIMP-2content and the expression of TGF-?1.Results:(1)Sang xing qing ying decoction could significantly reduce the coefficient of rat lung radiation pneumonitis(p<0.01),prevention group was superior to trea tment group, the three medicine groups were statistically different(p<0.05);lung pathology observations showed that prevention group, treatment group.western medicine group compared with model group,inflammatory cellinfiltration,alveolar congestion,exudation,bleeding,fracture spacing to a lesser extent.The lung tissue structure of blank group is clear.(2)Compared with the control group,model group,serum IL-6,TNF-? content value were significantly higher in eachtime(p<0.05).Compared with model grou p,prevention group, treatment group,western medicine group, serum IL-6,TNF-a, were significantly lower(p<0.05).(3)Compared with the control group,model group SOD activity in Lung tissue were decreased (p<0.05),MDA content increased(p<0.05);compared with the model group,the lung tissue of the medicine groups SOD activities increased,MDA content decrease.prevention group was superior to treatment group and western medicine group(p<0.05).(4)Compared with the blank group,2 to 4weeks,MMP-2,TIMP-2content in model group were increased(p<0.05), compared with model group, MMP-2,TIMP-2 content in prevention group,treatment group,western medicine group,was significantly lower(p<0.05).Conclusion:Sang xing qing ying decoction have a certain effecton the pre vention and treatment of radiation pneumonitis.The preventive effect maybe ach ieved through the following channels:enhance activity of SOD and reduce the MDA content; suppress serum levels of inflammatory cytokines IL-6,TNF-a,regu late the balance of MMP-2/TIMP-2,reduse the expression of TGF?1.
