Application Of PD-1 Antibody In Clinical Diagnosis And Immunotherapy Of Lung Cancer

BackgroundPD-1,as an important immune negative molecules involved in the immune regulation and stability of microenvironment.In cancer pathogenesis,often because of immune cells incompetence,lack of tumor cells to keep clear of in time,PD-1 antibodies can suppress the signal by blocking,partial immune cell function.This thesis mainly use the PD-1 antibody testing blood samples from lung cancer,and study the PD-1 antibodies effect of CIK cells kill tumor cell lung cancer.ObjectiveTo evaluate specificity and sensitivity of PD-1 antibody detection on the lung patients' blood samples,and to observe immune activity of the PD-1 antibody on killing tumor cells in vitro.MethodsPD-1 on lung cancer patients' PBMC membrane was detected by FCM,and soluble PD-1 in plasma was tested by ELISA.The Specificity and the sensitivity were calculated using ROC curve.After culturing CIKs,we tested killing assay about A549,H1299 cell lines,and primary cultured lung cancer cel s.Results1)PD-1s on lung cancer patients' PBMC(18.97%±2.32%)were higher than the health(11.10%±1.05%);advanced cancer patients' m PD-1s(23.07%±2.82%)were higher than earlier ones(12.81%±3.34%);lung cancer patients' s PD-1(1.66ng/ml±0.21ng/ ml)was less than the health(3.01ng/ml±0.36ng/ml);advanced cancer patients' s PD-1(1.14ng/ml± 0.18ng/ml)was less than the earlier(2.43ng/ml±0.34ng/ml).AUC of m PD-1 was more than 0.7,its sensitivity was 0.667 and the specificity was 0.867.2)The expression of PD-1 on CIK was 25.6%,the result of PD-1 antibody group changed from 31.2%±5.5% to 57.3%±8.1% on A549,from 34.4%±5.9 to 49.8%±5.7% on H1299,early lung cancer group kill rate increased from 35.5% plus or minus 4.5% of 59.0 + /-4.3%,middle-late stage lung cancer group kill rate from 31.8% plus or minus 4.0% to 54.0% plus or minus 4.9%.Conclusion This PD-1 antibody has a certain clinical diagnostic detection on lung cancer patients' m PD-1.When immune cells kill lung tumor cells in vitro,the PD-1 antibody could improve immune killing activity appropriately.
