Fulvic acid(FA)is one portion of humic acid which has complicated organic compound,the smallest molecular weight and the most medical value.The corrupt process,different sources and different process of extraction may be lead to the differences on chemistry and pharmacology.So it is necessary for us to make efforts on the conditions,for example source and purification,which have an affect on the treatment mechanism.In the past decades,a mass of studies had shown that FA has many properties such as anti-inflammatory,anti-bacterial,blood stasis,anti-ulcer and improving the immune function.However,the mechanisms of these effects have not clear.In 1980s,someone had excogitated that FA and sodium humic have effect on gastric ulcer in rat but they also did not explore the mechanism.In addition,how about the effect on gastric ulcer of young lignite fulvic acid is not be reported.Based on this,our study on the effect of gastric ulcer and its mechanism with young lignite-derived fulvic acid in Yunnan has a great significance.At the same condition,we draw the multiple samples from different regions to explore the influence on absolute ethanol induced gastric ulcer,H.pylori,xylol induced auricle swelling and RAW264.7 in mice.Comparing the results,we can conclude that the FA in different places has the differences of pharmacological activity.It provides some basic data and research for the development and utilization of young lignite in Yunnan.In this study,we use the green extraction technology which developed by our lab,through H2O2 oxidative degradation and 001x7 type cation exchange resin purifying method finally obtained the purified products from young brown coal of E'Shan,XunDian and ZhaoTong in Yunnan province.In the experiment of absolute ethanol induced gastric ulcer in mice,we measured the ulcer area?MDA?SOD?GSH and GSH-PX;The inhibition of H.pylori by disc method,this two experiments showed us the effect on gastric ulcer and the possible mechanism of FA from the three places.Additionally,the experiments of xylol induced auricle swelling in mice and LPS induced inflammatory on RAW264.7 to study the role in gastric ulcer and gastritis.The main contents and findings of this study are as follows:1.The FA of ES(50mg/kg)showed the dominant role in the inhibition of ulcer area?SOD?GSH and GSH-PX;The FA of XD(100mg/kg)had better effect on inhibition of ulcer area?MDA and GSH;The FA of ZT(50mg/kg?100mg/kg?200mg/kg)had the best effect on MDA and GSH in the tissue.The FA said above showed better effects on absolute ethanol induced gastric ulcer in mice than ranitidine(100mg/kg)as an positive drug.2.The result of bacteriostasis experiments by disc diffusion method on H.pylori revealed that the FA(50mg/mL)had no inhibited effect on H.pylori;the effect of FA(ES and XD)was enhanced with the increment of concentration;the FA of ZT almost had no inhibited effect on H.pylori.3.The pharmacological effects trend on xylol induced auricle swelling in mice of FA of ES and XD was similar.10mg/mL showed the best effect,5mg/mL is better and 20mg/mL is the worst.The FA of ZT(20mg/mL)is better than the FA of ES,as a whole,the FA of XD had the best effect on this experiment.4.The influence of FA on LPS stimulated RAW264.7 to produce NO:The FA of ZT had the best role in inhibited NO,the effect of FA of XD was better than ES.The effect was enhanced with the increment of concentration.The findings above suggest that fulvic acid from young lignite in Yunnan are not similar on absolute ethanol induced gastric ulcer,H.pylori,xylol induced auricle swelling and RAW264.7 in mice.On the whole,the FA of XD showed the best role in the study.The effect of FA on the relationship in gastric ulcer and gastritis remains to explore and perfect.This study further illustrated the research on fulvic acid should according to its regions and form;we also confirmed the mechanism of fulvic acid is completed with multi-factor.The development and utilization of Yunnan young lignite should according to the further study in order to avoid the unnecessary waste of resources.
