Objective To observe the insulin levels, the level of ventricular remodeling,the changes of adiponectin and adiponectin receptor 1 after insulin resistance in the wild and adiponectin gene knockout mice(APN- /-) mice,and to investigate the adiponectin on ventricular remodeling induced by insulin resistance.Methods Choosing eight male wild-type mice as insulin resistance group(IR),twenty-four male APN- /- mice were randomly divided into gene knockout groups(KO), gene knockout + insulin resistance group(KIR) and gene knockout + +insulin resistance + adiponectin intervention group(APN), each group has eight mice.The knockout group was given norma diet, the other three groups given high fat diet, APN group were injected intraperitoneally administered exogenous recombinant adiponectin(10 ug/kg. d).Detection test performed at 12 weeks.The mice were fasted 12 hours for one day in advance, taking the rat tail blood to measure rat tail fasting plasma glucose(FPG);measuring fasting serum insulin after taking(FINS), serum adiponectin(APN) and serum MMP 9 levels after taking serum,calculating insulin resistance index(HOME-IR), removeing the heart, then the left ventricle was isolated and said it,sweight, calculating left ventricular mass index calculation(LVWI). To observe the myocardial tissue morphology by HE staining and myocardial collagen by Masson staining in each group. Using Western Blot immunohistochemical method to test the expression of adiponectin and adiponectin receptor 1(Adip R1) protein in myocardium, Using real-time fluorescent quantitative PCR method to test the expression of myocardial Adip R1 m RNA.Doing Pearson linear correlation analysis in these indicators : LVWI and insulin resistance index, LVWI and serum adiponectin levels and expression of myocardial Adipo R1.Results(1)compared with IR group, the weight, FBG, FINS, HOMA- IR,Serum APN, myocardial adiponectin expression were significantly reduced in KO group; the FBG, serum APN, myocardial adiponectin and Adip R1 were significantly lower, whereas serum MMP-9 and LVW were significantly increased in KIR group; the FBG, FINS, HOMA- IR,serum MMP-9,LVWI and apoptotic rate were significantly reduced, but myocardial adiponectin and Adip R1 were significantly increased in APN group, the difference was statistically significant(P <0.05).(2) Compared with the KO group,the weight, FINS, HOMA-IR, serum MMP-9,LVWI and apoptotic rate were significantly increased, while the expression of myocardial APN and Adip R1 was decreased in KIR group; FINS and serum MMP-9 were significantly reduced,whereas the protein expression of APN,the protein and m RNA expression of Adip R1 was increased APN group, the difference was statistically significant(P < 0.05).(3)Compared with the KIR group, the FINS, HOMA-IR, serum MMP-9, LVWI and apoptotic rate were significantly reduced,but serum APN, the APN protein expression,the protein and m RNA expression of Adip R1 was increased APN group,the difference was statistically significant(P < 0.05).(4)From HE staining,myocardial cell arranged disorder in IR group;myocardial cell arranged disorder and nuclear stained uneven in KO group;KIR group myocardial cell disorder degree aggravating, even saw the muscle fiber fracture, deformation and other changes;myocardial arrangement neat, myocardial fibers disorder was improved in APN group.From Masson staining, blue collagen fibers increased,and myocardial fibrosis was significantly in IR group; interstitial collagen fiber segment myocardial bundles in blue in KO group;the degree of myocardial cells arranged disorder increased,part of the collagen fiber fusion in KIR group;the myocardial fibers were reduced significantly in APN group.(5)From Pearson linear correlation analysis, HOMA-IR HOMA-IR and the left ventricular mass index were positively correlated,and it negatively correlated with myocardial Adipo R1.Conclusion Insulin resistance can lead to ventricular remodeling.Exogenous adiponectin intervention can improve the IR induced ventricular remodeling...
