Objectives1. To analysis the association between the genetic polymorphisms in BRG1 and ARID1 B and the risk of Primary Liver Cancer.2. To explore the association between the genetic polymorphisms and the clinical characteristics of the Liver Cancer.MethodsWe conducted a case-control study, in shunde region of Guangdong province, to investigate the association of the genetic polymorphisms and the risk of Primary Liver Cancer.A comprehensive strategy was appliedto systematically explored the potential gene-gene and gene- environment interactions.Results1. The polymorphism(rs73013281)was significantly associated with the risk of Liver Cancer. As compared with the T allele, the CC homozygotes conferred an OR of 0.57(95% CI = 0.35-0.95; P = 0.030). The gene-environment interaction analysis with the PLCSignificant multiplicative and additive interaction were observed between rs73013281 polymorphisms and movement by crossover analysis(Pa=0.029,Pb=0.016). The interaction analysis indicated that there was a additive interaction between rs73013281 polymorphisms and the family history of tumor(Pa=0.040).In ARID1 B, there was a multiplicative interaction between haplotype and and smoking(Pb=0.032);Significant multiplicative and additive interaction were observed between haplotype and he family history of tumor(Pa=0.014,Pb=0.013).2. For the progression of liver cancer, the onset age was significantly association with TNM stage and portal vein tumor. Additionally, the individuals with the rs11085752 CC genotype in BRG1 showed an earlier average age of onset, as compared with those carrying the A allele.Conclusions1.Thers73013281 variant in ARID1 Bcould interaction between movement and family history oftimor affecting the Shunde crowd PLC susceptibility;ARID1B haplotype associated with smoking and family history of tumor of interaction liver cancer have a major impact in Shunde.2.The earlier onset age could increase the metastasis risk of liver cancer, while the rs11085752 variant in BRG1 was significantly associated with the onset age. Further studies with large sample size are warranted to validate these results.
