Objective To investigate the role of aquaporin1(AQP1) in pathologic process of lung injury in mouse models of fat embolism syndrome(FES).Methods A total of 96 healthy male C57BL/6J mice were assigned to control group and FES group which was grouped by 4,6,12,24,and 48 hours with 16 mice per group,according to the random number table.Allogeneic perinephric fat was injected to rat caudal veins in FES groups while the control group was given the same volume of physiological saline.Lung samples were obtained from each group W/D to verify the FES models and observe the pathologic process.Expression of AQP1 was detected by westernblot and immunohistochemistry.Additional 24 C57BL/6J mice were divided into control group, FES 24 hour group,and AQP1 inhibitor group according to the random number table,with 8 mice per group.Pathologic process of FES—induced lung injury was detected after the inhibition of AQP1.Results: The construction of lungs in FES group have been damaged more seriously compared with the contrast group, The values of W/D and the expression of AQP1 were significantly higher than the contrast group.At the same time, inhibition of AQP1 can decrease the damage of lung injury.
