Abstract/Summary:
Objective: To establish the lung injury model of mice induced by fat embolismsyndrome (FES), and investigate the expression of AQP4in it.Methods:1120healthy male C57BL/6J mice were evenly and randomly assigned intocontrol group and FES groups (1-5group). FES groups were injected with allogeneicperinephric fat through caudal veins. After4h,6h,12h,24h and48h of injection,pathological morphology, W/D ratio as well as serum IL-6level were be examinedrespectively to ensure the success of the model and observe the pathology process.Followed the expression of AQP4was detected by western blot, RT-qPCR andimmunohistochemistry.256healthy male C57BL/6J mice were evenly and randomlydivided into control group, DMSO group, FES group and inhibitor group, in order toobserve the changes in the pathology process of FES induced lung injury after inhibitingAQP4.Results: Compared with the control group, the lung tissue of FES groups has moreserious damage. The value of W/D and the expression of AQP4were significantly higherthan the control group P<0.05. Meanwhile, inhibition of AQP4may decrease thedamage of lung injury.Conclusions: AQP4may be involved in the pathology process of FES induced lunginjury.
