Objective The problem of bone defect and nonunion repair has been very difficult,and many methods have been tried.Recently researchers have designed biomimetic bone matrix material,in order to solve the problem of bone defect and nonunion of the knotty problem provides a new method to repair,this paper not only effectively utilizes three calcium phosphate good biodegradability and morphology characteristics as well as the carrier of the function,but also make full use of the type? collagen cells and good biocompatibility,synergistic effect produced by the combination and both,especially the bone morphogenetic protein can induce animal or human mesenchymal cells differentiate into bone,cartilage,ligament,tendon and nerve tissue,and then using plasmid DNA as a gene carrier,made with the occurrence of bone morphogenetic protein-2 as a bone repair material to form plasmid wrapping the beta gene three calcium phosphate solution type /? collagen scaffold materials,and this observation by experimental study in vitro The physical and chemical properties,biocompatibility and its ability to induce and conduct the bone formation of the composite materials are highly effective for the reconstructi on of bone tissue.Methods The preparation of nano porous-TCP/ collagen scaffold and the loading of plasmid DNA containing the target gene BMP2 was used to form the gene modified scaffold materials.In vitro culture system of composite scaffold material and MC3T3-E1 cell line of mouse bone.Which can be divided into stent group(Z)and plate group(M),according to the different concentration of eachsubdivided plasmid into 2 groups.The cell proliferation was detected by scanning electron microscope and light microscope,and cell proliferation was detected by Blue Alamar method.Cell cycle was detected by Analyzer MuseTMCell method.The proliferation of Cdk4 and Cdk2 were detected by real-time fluorescence quantitative PCR.Test results were statistically analyzed.Results Containing human bone morphogenetic protein 2 for target gene of plasmid DNA modified nano tricalcium phosphate calcium ?-type collagen solution composite surface above have large size pores.This study shows composite materials and simple hBMP-2 as compared to that of the target gene of plasmid DNA,can promote the former into osteoblast adhesion,proliferation and metabolism,showed that the pre osteoblasts has good cell compatibility.Conclusion The human bone morphogenetic protein-2 is the target gene of plasmid DNA modified nano-beta-phosphate three calcium type /? collagen solution compo site material has the role of promoting the proliferation of mouse bone cells.Containing human bone morphogenetic protein-2 gene of plasmid DNA modified nano tricalcium phosphate calcium I-type collagen solution composite of preosteoblast cell proliferation were superior to the simple containing human bone morphogenetic protein 2 gene of plasmid DNA and pure nano beta phosphorous acid calcium / type-? collagen solution of scaffold materials.
