Objective: Oral squamous cell carcinoma is the most common malifnant tumor of oral and maxillofacial region,serious damage to human health.This experiment to study the influence of exosomes secreted from oral squamous cell carcinoma cells(OSC-3、OSC-4) on their malignant cellular potential.Methods:We cultivate two kinds of oral squamous cell carcinoma cell lines: OSC-3 and the OSC-4 cells, then separate and purify exosomes from the medium,and normal OSC-3 and OSC-4 as control groups, OSC-3 and the OSC-4 with exosomes as experimental groups.By using gold-labeled immune chromatography, we find that CD9 is expressed on the membranes of the exosomes.then we detect the effects of the exosomes on the OSC-3 and OSC-4 cell proliferation by MTT assay; by cell scratch experiments to evaluate the ability of cell migration;the transwill chamber invasion experiment to detect the influence of the exosomes on the cell invasive ability.Results: Gold-labeled immune chromatography shows that CD9, the separation and purification which content is the exosomes; MTT test results showed that after EXO funtion the OSC-3 and OSC-4 proliferation ability in group 24h、48h、72h all enhancement(P<0.05),scratches healing experimental results showed that OSC-3 and OSC-4 migration ability enhancement after exosomes(P<0.05);transwill chamber attacking experimental results showed that in OSC-3 and OSC-4 after EXO function the number of extra cellular body cells stained significantly more than the control group,after statistical analysis, the number of invasive cells was significantly more than the control group,invasion ability of OSC-3 and OSC-4 also enhance(P<0.05).Conclusion: The exosomes secreted from oral squamous cell cancer cells can promote the abilities of cell proliferation、migration and invasion.
