Objective: This study collected from normal full-term pregnant women at normal expectant, normal latent and active phases of the first stage, the first 24 hours after birth and the 72 hours after labor, peripheral blood specimens, five time points to determine the concentration of serum lipid oxygen element, and establish the animal model to further explore the significant role of lipoxin A4 in childbirth endometrial lesion and involution.Set up six animal experimental groups, rats in the experimental groups continuous injected i.p. with Boc-2, LXA, LPS on GD 18,GD 19 and GD 20, whereas rats in control group were injected i.p. with equal vehicle(saline), in order to explore the role of lipoxin A4 in the process of childbirth.Material and Method: Application of RT-PCR, western blot test methods, such as validation of Lipoxygenase enzymes LOX-5, LOX-12 and LOX-15 m RNA and protein expression in different times, observe the inflammatory factors, such as the concentration of IL-6, IL-8 and the relationship between the changes and inflammatory cells infiltration.Observing the endometrium inflammatory cells infiltration through the different time points of endometrium for neutrophils and macrophages by immunohistochemical staining.Result: The result reveals that lipoxin A4 really participated in the inflammation control in the process of childbirth, but did not play a vital role for the delivery. Through the observation and study of postpartum uterus tissue found inflammatory cells also involved in the process of uterine instauration, especially neutrophils and monocytes, and the concentration of lipoxin A4 has the corresponding changes in trends, hints of endometrial repair as a dynamic process of inflammation, lipoxin A4 plays an important role in promoting endometrial involution and maintain local inflammatory cells and inflammatory factors homeostasis. Accordingly, we conclude that: lipoxin A4 not play a crucial role in the delivery and launch the progress of labor, but do plays an important effect on the promotion of postpartum endometrial repair,involution, and control the local inflammatory microenvironment.
