Objective To investigate the expression of Kinesin 2a(Kif2a), Matrix Metalloproteinase-2(MMP-2) in lung squamous cell carcinoma, and to explore their relationship to the clinical pathological characters and correlation between them. Methods The expression of Kif2 a and MMP-2 was detected immunohistochemistrically in 60 cases of lung squamous cell cancinorma and 28 cases of pericancerous normal lung tissue. Results The expression of Kif2 a was found in nucleus and/or cytoplasm, and MMP-2 only in cytoplasm. The positive rate of Kif2 a and MMP-2 in lung squamous cell cancinorma were both significantly higher than that in the normal tissue(X2=21.614?26.110, P < 0.01), and there were also significant difference in the positive intensity(Z=-4.981?-5.25,P<0.01).The positive expression of Kif2 a and MMP-2 in lung squamous cell carcinoma was closely correlated with the cancer TNM staging and lymph node metastasis(Z=-2.046 ~-2.214,P<0.05), but not correlated with the sex, age and size(P>0.05). Furthermore, the expression of MMP-2 was also correlated with differentiation(Z=-2.078,P < 0.05),and that of Kif2 a was not correlated with differentiation(P >0.05).The expression of Kif2 a and MMP-2 was positively correlated(r=0.468,P<0.01).Conclusion(1)Both the positive rate and intensity of Kif2 a and MMP-2 are different between carcinoma and pericancerous normal lung tissue, it indicates that the over-expression of Kif2 a and MMP-2 may relate to occurrence of lung squamous cell cancinorma.(2)Both Kif2 a and MMP-2 express strong either in stage?~? or with lymph node metastasis reveal that they may also play important roles in tumor progression and metastasis.(3)Positive correlation is found between Kif2 a and MMP-2 in lung squamous cell cancinorma suggest that over-expression of Kif2 a and MMP-2 may has synergetic effect in the process of occurrence and development of lung squamous cell cancinorma.
