Objects A comparative analysis of PA group,PP group and Normal group HIF-1?protein and mRNA expression levels in the placenta tissue, to explore the possible role of HIF-1? in morbidly adherent placenta.Methods we recruited 60 pregnancies from Fujian Maternity and Child Health Hospital between January 2015 and November 2015,by clinical manifestations,ultrasonic examination and postoperative pathological diagnosis of placenta accreta,10 patients,collecting the accreta site tissue and the normal site;the clinical manifestation and the ultrasound diagnosis of placenta previa,20 patients,collecting central site and the marginal site tissue;we recruited 30 pregnancy by cesarean section because of abnormal birth canal, abnormal fetal position or social factors as control group, collecting the umbilical cord attached to the corresponding area of the placental tissue.ELISA, RTQ-PCR techniques were used to detect the PA group, PP group and Normal group in the placenta tissue HIF-1? protein and mRNA expression of HIF-1?.Results1.Each maternal clinical data analysis: PA group, PP group and Normal group maternal age, occupation, times of cesarean delivery, since the uterus surgery,there were no statistically significant difference(P>0.05),and data were comparable; PA group maternal hospitalization days, postoperative blood loss were significantly higher than that of Normal group(P < 0.05), but PA group and PP, PP group and Normal group there was no significant difference(P > 0.05); PA group maternal hospitalization expenses, the number of pregnancy, intraoperative blood loss, operating time were significantly higher than PP group and Normal group(P< 0.05), PP group and Normal group there were no significant difference(P > 0.05);PA, PP group maternal level of education, gestational age, hospital childbirth mode(Emergency, outpatient, hospital referral) compared with Normal group difference were statistically significant(P < 0.05).2.Analysis of clinical data in 3 groups of perinatals: PA, PP group and Normal group neonatal apgar score difference had no statistical significance(P > 0.05); PA,PP group, the placenta weight, Neonatal preterm birth rates,neonatal birth weight and the rate of neonatal checking in NICU or high view room or neonatology department to have significant difference compared with Normal group(P < 0.05),while PA, PP group was no significant difference(P > 0.05).3.PA group, PP group and Normal group in the placenta tissue were detected HIF-1? protein expression, PA group, PP group expression level was significantly higher than in Normal group(P < 0.05), and expression level of PA group was higher than PP group, the difference was statistically significant(P < 0.05).4.PA group, PP group and Normal group in the placenta tissue were detected HIF-1? mRNA expression, PA group, PP group expression level was significantly higher than in Normal group(P < 0.05), and expression level of PA group was higher than PP group, the difference was statistically significant(P < 0.05).4.the HIF-1? protein expression differenc in accreta site and normal site of PA group was no statistical significance(P > 0.05), the HIF-1? mRNA expression differenc in accreta site and normal site of PA group was no statistical significance(P > 0.05).6.HIF-1? protein expression level difference in the central site and marginal site tissue of PP group was not significant(P > 0.05),HIF-1? mRNA expression level difference in the central site and marginal site tissue of PP group was not significant(P > 0.05).Conclusion1.MAP could be related with Perinatal adverse pregnancy closely.2.The expression levels of HIF-1? in placenta may play an inportant role in pathogenesis of MAP.3.Thc expression of HIF-1? in placental tissues in patients with MAP was significantly higher than normal,which could Serves as an important indicator for the early diagnosis of MAP.
