BACKGROUND AND PURPOSE:Facing the increasingly serious problem of the infection following orthopedics surgery with implant, to modify and to load antibiotics in the titanium plate surface have become the research hotpot in the field of orthopedics.This research adopts the anodic oxidation method to prepare the Ti O2 nanotube coating on orthopedic titanium plates,and the electrodeposition method to load iodine on the nanotube. By adjusting the parameters in the experiments, this research studies the characterization of nanotubes contained iodine,and the antibacterial properties of iodine-supported titanium implant under the laboratory conditions, plans to develop a kind of orthopaedic implants with antibacterial properties.Methods:Firstly, prepare Ti O2 nanotubes by electrochemical anodization, with the fluorine-containing glycerol as the electrolyte, a polished, passivated pretreatment orthopedic titanium plate as the anode,and a platinum plate as the cathode. The interval between the two electrodes is 2cm, the temperature of the electrolyte is 25 ?, and the speed is 130 beats / min. After anodized 24 h under the voltage of 40 v, Ti O2 nanotubes should be ultrasonic cleaned and dried;Secondly, Electrodeposition method is adopted to load iodine. Prepare 1000 ppm KI aqueous solution?1ppm = 1mg / kg = 1mg / L?, which contains 76.50 percent iodine,adding an appropriate amount of iodine solution. With the prepared Ti O2 coating nanotubes used as an anode and the platinum plate as a cathode, the electrodeposition is taken under voltage of 65 v in 30 min. Then Ti O2 nanotubes is washed with ethanol and air-dried. Using scanning electron microscopy?sem? to observe nanotubes, X-ray photoelectron spectrometer to analyse the element on the surface of the nanotubes;Thirdly, group the objects. the ten titanium plates with iodine coating is regarded as group A, and the ten untreated titanium plates group B. The same amount of colonies were inoculated into the two groups, incubated at 37 ? for 6 hours. Then, wash the two groups with 5ml PBS buffer and after diluting the cleaning fluid 50 times. Take50 ul diluted fluid, smear on agar medium and culture in 37 ? for 24 h. Then observe and counted the number of colonies.Result:1. With the diameter of about 100-200 nm and the length about 1um, the nanotubes prepared by the 24 h anodic oxidation presents a regular shape;2. Scanned by electron microscope, the nanotubes which load iodine by electrodeposition method have polymer particulate iodine on the surface. The amount of iodine is about 5% analyzed by X-ray fluorescence spectroscopy;3. The differences of the amount of colonies between the 24 h cultured group A and group B is statistically significant?P <0.05?Conclusion:Electrochemical anodic oxidation method is a simple and scientific methods which can produce nanotubes with uniform configuration. The nanotubes anodizing for 24 h after has a suitable length and diameter, which can be used as an excellent drug load carrier.The successful load of iodine is proved by the X-ray fluorescence spectroscopy elemental analysis, semi-quantitative determination The antibacterial experiments show that under laboratory conditions, compared with the regular ones, the titanium plates with iodine coating prepared in this project has a stronger antimicrobial properties.
