Objective: Study the effect of Ang II and telmisartan playing in the progression of prostate cancer and its possible mechanism.Methods: Using Western blotting and q RT-PCR method to detect AT-1 expression in LNCa P, DU145 and PC3 cell lines.Using tissue microarray to detect AT-1 expression in different histological types of prostate tissues(normal prostate, prostate hyperplasia, prostate cancer). Detect the effect of Ang II and telmisartan(AT-1 receptor antagonist) on proliferation of cancer cells by CCK-8 method. Using transwell method to detect the effect of Ang II and telmisartan on migration and invasion of cancer cells. Detect the effect of Ang II and telmisartan on MMP-2, MMP-9 expression of cancer cells via Western blotting, which is related on invasion.Results: AT-1 is expressed in the three prostate cancer cell.From high to low of the degree of expression is DU145,PC3 and LNCa P. The results of tissue microarray suggest AT-1 expression increase with the development of prostate cancer. Ang II can promote the proliferation, migration and invasion of DU145, and telmisartan can inhibit it. Ang II can enhance the expression of MMP-2 of DU145 associated with invasion and telmisartan can inhibit the effect.But they have no obvious effect on the expression of MMP-9.Conclusion: Ang II and telmisartan play a certain role in prostate cancer progression. In the treatment of prostate cancer,telmisartan may play a certain role, which needs further study.
