Objective Through the best preparation method, using the optimal formulation preparation of Chinese yam polysaccharide liposomes. The effects of physical and chemical properties of liposomes, which incluces morphology, size distribution, Zeta potential and stability. The preparation of freeze-dried powder and evaluate product stability. Through the analysis method to evaluate sustained-release effect of in vivo and in vitro. To explore Chinese yam polysaccharide liposomes influence on acute liver injury in rats, provide theoretical basis for the efficacy of yam polysaccharide.Methods The preparation of Chinese yam polysaccharide liposomes use the method of reverse evaporation and film extrusion. Entrapment efficiency as the evaluation index. On the basis of single factor, using orthogonal experiment design to screen the optimal prescription of the preparation of the yam polysaccharide liposomes.The encapsulation efficiency was determined by the method of low temperature and high speed centrifugation, and then calculate the drug loading efficiency.particle size, morphology and Zeta potential of Chinese yam polysaccharide liposomes are characterized separatedly by the Nano-ZS90 type laser nanometer size. Use the Hitachi7700 type transmission electron microscope to observe morphology of Chinese yam polysaccharide liposomes. The dialysis method is used to determine in vitro release, This experiment adopted four kinds of equations including zero order, first order, Higuchi and Hixcon-Crowell, to select the curve fitting equation of the Chinese yam polysaccharide liposomes.By appearance, color and dispersion as evaluation indicators, to screen of freeze-dried protective agent freeze. Freeze dried powder sterilization method was selected with the encapsulation efficiency as the index. Rats were randomly divided into normal control group, model group, positive control group, high, medium and low concentration group of yam polysaccharide liposomes. Each group is made up of 10 rats. Except the normal control group, Each group of rats by intraperitoneal injection of carbon tetrachloride solution preparation of liver injury model.After24 h,rats would be given yam polysaccharide liposomes and positive drug by tail vein.After 7 days, abdominal aorta in blood, to determine the AST, ALT, ALP activity. Samples from the liver supematants tested for MDA,T-SOD and GSH content.collecting the right lobe of the liver to test,though hematoxylin-eosin staining, then observe the liver damage degree.Results According to the optimal prescription, chinese yam polysaccharide liposomes were prepared. Its entrapment efficiency was(83.29 + 2.13) %, the average drug loadings was(3.83 + 0.11) %, the average particle size was(121.6 ± 2.7) nm, PDI was 0.136 ±0.011, Zeta potential was(-31.8 ± 0.69) mV. Raw medicine of Chinese yam polysaccharide released completely after 6 h, yam polysaccharide liposomes release rate was 21% after 6h, the accumulative release rate was 77.48% after 36 h. Concentration of 5% mannitol have the best protection effect to freeze-dried powder. Compared with the normal group, model rats showed significantly higer liver index,the levels of AST, ALT and AKP were significantly increased in the blood, Compared with normal group, model group rats developed markedly liver damage, accompanied with an increased level of MDA, and decreased level of T- SOD and GSH, and there was significant difference(P?0.01).Compared with model group, when given different doses of yam polysaccharide liposomes and positive drugs after injection, the levels of AST, ALT and AKP activity decreased, existing the extremely significant difference(P?0.01).High concentrations of yam polysaccharide liposomes group inhibition activity of AST slightly stronger than the positive drug group, but no significant difference(P = 0.631). Compared with control model group, accompanied with decreased level of MDA, and increased level of T-SOD and GSH in liver tissue homogenate, but on GSH index between high, medium and low concentration group of yam polysaccharide liposomes were no significant(P>0.05). The pathological sections of liver showed that the liver structural of model rats are destroyed, appeared hepatocyte fatty degeneration, cellular inside the fat vacuole were abvious and there are many focal necrotic areas. when given different Concentrations of yam polysaccharide liposomes and positive drugs after injection,high concentrations of yam polysaccharide liposomes group and positive control group is the most significant effect on the liver tissue repair damaged.Conclusions The Chinese yam polysaccharide liposomes were prepared by the reverse evaporation- film extrusion method, which uniform particle size distribution, coating rate is higher, the stability is good, have sustained-release effect in vitro. In vivo animal experiment results show that the yam polysaccharide liposomes can repair the damaged liver cell, on acute liver injury caused by carbon tetrachloride has a certain resistance.
