Objective: to detect the expressiones of IL-8 and its receptor IL-8 RA and IL-8 RB in Hela cervical cancer cells,compare the expression difference of IL-8 between cervical cancer tissues and normal cervical tissues,and to explore the effects and mechanisms of exogenous IL-8 on carcinogenic potential of Hela cells.Methods: The expression of IL-8 and its receptor IL-8 RA and IL-8 RB in Hela cells were detected by RT-PCR and Western Blot;The expression of IL-8 in a cervical tissue microarray was explored by immunohistochemistry;The effects of exogenous IL-8 on growth and migration of Hela cells were tested by CCK8 and Transwell assays,respectively;The expression of genes regulated by IL-8 was detected by RT-PCR.Results: IL-8 and its receptor IL-8 RA and IL-8 RB were expressed in Hela cells,the relative expression of L-8RA and IL-8RB mRNA were 4264.67±505.80 and1644.33±320.72 respectively,there was significant difference between IL-8RA and IL-8RB mRNA expression(P<0.01);The relative protein expressiones of IL-8RA and IL-8RB were 0.86±0.08 and 0.50±0.05 respectively,there was significant differences between L-8RA and IL-8RB protein levels(P<0.01);The expression scores for IL-8 in cervical cancer tissues and normal cervical tissues were respectively 8.13±3.53 and 3.86±2.10,there was significant differences between the two groups(P<0.05).The growth rates(absorbance values)of 0 ng/ml?20 ng/ml?40 ng/ml?60 ng/ml and 80 ng/ml IL-8 were respectively 1.08±0.12 ? 1.12±0.14 ? 1.28±0.04 ? 1.34±0.06 and 1.27±0.06,40 ng/ml?60 ng/ml and 80 ng/ml IL-8 can significantly promote proliferation of Hela cells as compared with 0 ng/ml IL-8(P<0.05);The number of migration cells in the groups of 0 ng/ml?20 ng/ml?40 ng/ml?60 ng/ml and 80 ng/ml IL-8 were respectively 58.17±8.82?58.83±10.32?76.67±9.58?83.50±5.68 and 72.00±6.48,40 ng/ml?60 ng/ml and 80 ng/ml IL-8 can significantly promote migration of Hela cells as compared with 0 ng/ml IL-8(P<0.05);Compared with 0 ng/ml IL-8,40ng/ml and 60ng/ml,IL-8 can up regulate the expressiones of IL-8RA and IL-8RB genes,down regulate the expressiones of ERK and NUMB genes(P<0.05).Conclusions: The expression of IL-8 in cervical cancer tissues is significantly higher than normal cervical tissues,exogenous IL-8 can promote the carcinogenic potential of Hela cells by enhancing the expressiones of its receptor IL-8RA and IL-8RB,down regulating the expression of ERK and NUMB.
