Objectives Using immunohistochemical method to study the expression of 5-HT? 5-HT3 R in intestinal mucosa of the diarrhea prominent irritable bowel syndrome. so as to provide basis for the study of the pathophysiology of the diarrhea prominent irritable bowel syndrome and provide molecular targets for treatment.Means 1 From September 2014 to June 2015, We select 40 patients in medical clinic in the affiliated hosipital of Science and Technology of the North China University who has IBS-D and see a doctor as the case group. The patients into the case group must be conformed to the criteria of IBS-D of the Rome?. We select 24 cases without symptoms of digestion of healthy volunteers and medical examinations as a control. 2 Before colonoscopy, the subjects should read into applications of the groups and digestive endoscopy agrrement. Before the colonoscopy examination, subjects should sign the relevant document. Into the mirror of the ileum, we take a biopsy of intestinal mucosal tissue. We observe the intestinal mucosa in detail while returning the mirror. When the mirror back to straight-sigmoid colon junction, we take a biopsy of intestinal mucosal tissue. The biopsy tissue fixed quickly with formablehyde solution. Tissues Embedded with paraffin, then tissues maked into scile. All the section stained with HE to observe the intestinal mucosa. The expression of 5-HT, 5-HT3 R in intestinal mucosa were observed by immunohistochemical method, and the expressions intensity were measured by integral optical density. 3 SPSS19.0 was applied to the experimental data for statistical analysis and processing. Before statistical analysised, all experimental data were f test and normal distribution test. Measurement data was repersented by mean standard±deviation. Chisquare test be used to compare the difference between the count data. T test be used to compare the difference between the two independent samples. We considered P ? 0.05 statistically significant, and P?0.01 statistically significant evidently.Results 1 The average IODs of 5-HT in rectum-sigmoid junction parts of intestinal mucosa in case group were higher than those in the control group? 98.95±64.86vs59.21±40.49, t=-2.698, P=0.009??P ? 0.01?, the difference was statistically significant observably; The average IODs of 5-HT3 R in rectum-sigmoid junction parts of intestinal mucosa in case group were higher than those in the control group? 78.85±37.38vs34.83±15.44, t=-5.480, P=0.000??P ? 0.01?, the difference was statistically significant observably. While the average IODs of 5-HT in terminal ileum had no statistical difference compared with the control group?59.21±40.49vs55.82±32.78, t=-1.424, P=0.159??P>0.05?; The average IODs of 5-HT3 R in terminal ileum had no statistical difference compared with the control group?40.32±14.59vs34.84±15.43, t=-0.367, P=0.715??P>0.05?. 2 5-HT immunohistochemical expression in the case group in rectum-sigmoid junction mucosa of patients had statistical difference than it in terminal ileum mucosa? 98.95±64.86vs59.21±40.49, t=-8.041, P=0.000??P ? 0.01?; 5-HT3 R immunohistochemical expression in the case group in rectum-sigmoid junction mucosa of patients had statistical difference than it in terminal ileum mucosa? 78.85±37.38vs40.32±14.59, t=-8.091, P=0.000??P ? 0.01?. 5-HT ? 5-HT3 R immunohistochemical expression in the control group in rectum-sigmoid junction mucosa of patients had no statistical difference than it in terminal ileum mucosa. 3 In this study,we found that the IODs of 5-HT in the case group of female patients were higher than male patients?99.67±65.93vs84.32±40.95, t=-2.397, P=0.000??P?0.01?, the difference was statistically significant observably; The IODs of 5-HT3 R in the case group of female patients were higher than male patients?76.82±32.16vs58.25±30.08, t=-2.950, P=0.000??P?0.01?, the difference was statistically significant observably.Conclusions 1 The expression of 5-HT?5-HT3 R in rectum-sigmoid junction colon of IBS-D patients is enhanced. 2 The expression of the 5-HT?5-HT3 R in rectum-sigmoid junction colon of IBS-D pations is higher than other parts of the intestinal tract. 3 The expression of 5-HT?5-HT3 R of Patients with IBS-D may be gender differences, of which express enhancement for the female patients.
