ObjectiveInvestigate the relative expression level of Notch3 in different pancreatic cancer cell lines, and its expression correlated with the expression of VEGF, cell proliferation and cell migration, and to provide important evidence for targeted therapy and early diagnosis for pancreatic cancer. MethodsWe detected the expression of Notch3 in HPDEC and several pancreatic cancer cell lines(Bx PC-3, PANC-1, As PC-1, Capan-1, Capan-2) by the methods of q RT-PCR finding that the expression of Notch3 in Capan-1 and Capan-2 is much higher than other cell lines. Then we transfected these two cell lines with Lipofetamine 2000 reagent using either negative control or target si RNAs. After that we detected the expression of notch3 and VEGF by Western Blot, detected the cell vitality by MTT, and the migration ability of the cells by Wound-Healing Assay. Data statistical analysis was performed with SPASS17.0 software. ResultsThe results turned out that the expression of notch3 and VEGF were both reduced in silenced groups, and also the cell vitality and cell migration ability were both receded in silenced groups. ConclusionsThis research revealed that the expression of notch3 and VEGF are correlated in pancreatic cancer, suggesting that notch3 may plays an important role in the VEGF associated vascular angiogenesis in pancreatic cancer. And this research also proved that notch3 improves the cell proliferation and cell migration in pancreatic cancer, suggesting that notch3 is related to the advance of pancreatic cancer and it's malignant degree.
