Objectives:The primary aim of this study was to evaluate the antimicrobial activity of Gutta Flow 2 compared with AH Plus, Real Seal and Pro Root MTA against Enterococcus feacalis, Escherichia coli and Candida albicans by means of both agar diffusion test(ADT) and modified direct contact test(DCT).Methods:The ADT was conducted on double-layer plates. Both fresh and 1-day-setting sealers were measured. For DCT, a 20 μL microorganism suspension was exposed to the sealers for 10, 30 and 60 minutes, which were freshly mixed or allowed to set for 1 or 7 days. Then 180 μL fresh media was added and survival of microorganism was determined by using 10-fold serial dilution. After incubation for 48 h, colony-forming units(CFU) were calculated and their log10 values were converted. The data were analyzed by using ANOVA and Tukey tests(a = 0.05)Results:Gutta Flow 2 did not exhibit any antimicrobial activity in both ADT and DCT. In ADT, fresh Real Seal had the largest inhibition zone against all the tested microbes, followed by AH Plus and Pro Root MTA. In the set group, only Pro Root MTA demonstrated inhibition zones. In DCT, fresh AH Plus eradicated all the three tested microorganisms, while its antimicrobial activity diminished significantly with time. The antimicrobial effect of Pro Root MTA and Real Seal were relatively stable, which lasted for 1 and 7days respectively.Conclusion : Gutta Flow 2 had no antimicrobial activity. AH Plus only demonstrated immediate antimicrobial efficacy while delayed antimicrobial properties were detected in Pro Root MTA and Real Seal.
