Objective:To systematically assess the efficacy of acotiamide in the treatment of functional dyspepsia(FD).METHODS:Studies on acotiamide in the treatment of FD were collected and re viewed through searching Pub Med,Embase,the Cochrane Central Register of Contr lled Trials,CBM,the Chinese Academic Journals FUll-text Database and Chinese VIP Full-text Database.The Revman5.3 software provided by the Cochrane Collab oration was used for assessment.RESULTS: A mata-analysis was conducted on 1442 patients from 5 randomized controlled trials with respect to the usage of acotiamide,which divided into treatment group(n=721) and control group(n=721).The results of heterogeneity showed that?2=3.42,P=0.49,I2=0%,therefor the fixed effect model was selected.The meta-analysis shows that treatment group were significantly different from control group(RR=1.39,95%CI [1.24,1.56],P<0.05).The subgroup analysis indicated that there was no statistical significant between treatment group and control group after a treatment of 2weeks(RR=2.00,95%CI [0.57,6.96],P=0.28),however the difference had statistical significant after a course of 4 weeks treatment(RR=1.39,95%CI [1.24,1.556],P<0.05).In adverse reaction,triglycerides increased?alanine aminotransferase increased??-glutamyltransferase increased?serum bilirubin increased?serum prolactin increased?white blood cell count increased?diarrhoea?nasopharyngitis implied no statistical significant among treatment group and control group except constipation?CONCLUSION: As a novel gastroprokinetic agent,the usage of acotiamide in th e treatment of FD can obviously relief the global symptom of FD,which suggests a better therapeutic effect than placebo,futhermore a course of 4 weeks treatment was more effective than 2 weeks.but adverse reaction followed up should be valued.However more high quality trails are needed for further confirmation.
