Objective:Periodontal disease is common oral disease in adults, but the incidence of periodontal disease in children and adolescence is constantly increasing. The initiating factor is plaque biofilm, where Porphyromonas gingivalis(P.gingivals), the main pathogens of periodontitis, play an important role in the destruction of periodontal tissues. Erythritol is a safe, non-toxic side effect, low calorie value, non-cariogenic food additives, and intended for use primarily as a new sweetener added to foods, winning the people's favorite. Currently, scholars have focused on the anti-caries of erythritol and founded that it can suppress the growth, acid generation and adhesion of cariogenic bacteria which of Streptococcus mutans(S. mutans) is the most important pathogenic factors of the pit and fissure caries.The aim is to research the inhibition effect of erythritol on growth and adherence of Porphyromonas gingivalis(P.gingivals), and to further study the influence of erythritol on the activity of trypsin-like protease(TLP) generated by P.gingivals, and to provide experimental evidence and new thinking of the erythritol on the prophylaxis and treatment of gingival diseases and periodontitis.Methods:In this study, P.gingivalis ATCC33277 was selected as the experimental bacterial strain. P.gingivals was cultured respectively in brain heart infusion broth(BHI) including 2%, 6%, 10%, 15%, 20% erythritol under anaerobic conditions(80%N2, 10%H2, 10%CO2) at 37?. The optical density(OD) values were measured at 4-h intervals and the growth curves were plotted. The number and color change of bacteria colonies were observed by using coated plate count method. The cementum blocks of the orthodontic extraction premolar were used for biofilm carrier of P.gingivalis to observe attachment effect of P.gingivalis cell by using the Scanning Electron Microscope(SEM) and the Confocal Laser Scanning Microscop(CLSM). After 48 h of incubation at 37? under anaerobic conditions, the culture supernatants, disrupted cell suspension,and the arginine protease(Rgp) were collected. And then survey and evaluate the vitality of TLP and Rgp.Results:The experimental results indicated that erythritol in a concentration-dependent mode was capable of restraining the growth and adhesion of P.gingivalis, the higher the concentration the better inhibition(P<0.05). Erythritol extended the time of color change of P.gingivalis from white to black by gathering hemoglobin in the medium. Erythritol also inhibited the activity of TLP and Rgp produced by P.gingivalis(P<0.05), and the influence on TLP in the cell membrane was better than the freestate.Conclusion:The conclusion is that erythritol inhibited the growth and adhesion of P.gingivalis, also suppressed the activity of TLP produced by P.gingivalis, the higher the concentration the better inhibition.
