Objective:(1)To utilize improved Allen's weight drop method to establish the SD rat model of spinal cord injury, evaluate its feasibility;(2)To investigate effect of ganglioside combine with glucocorticoids pluse therapy on the NGF expression after spinal cord injury in rats by means of immunohistochemical and fluorescent quantitative PCR methods;( 3) To evaluate the influence of glucocorticoids pluse therapy on neural functional recovery of spinal cord injury in rats.Method : Two hundred and forty Sprague-Dawley rats were randomly divided into four groups by random number table method : group A,model contral group(n=60),the rats were treated by the tenth thoracic laminectomy;the rats of group B?C?D were were made as SCI models by improved Allen's weight drop method in T10 level after thoracic laminectomy;group B,SCI group(n=60),the rats were administrated amount of saline by the caudal vein injection; group C, GM1 group( n = 60),the rats were administrated ganglioside GM1(10mg/Kg/d) by the caudal vein injection; group D,GM1 +MP group(n=60),the rats were administrated ganglioside GM1(10mg/Kg/d)and Glucocorticoids Pluse Therapy(30mg/kg within the first hours,and then 5.4mg/kg/h within the next 23 hours) by the caudal vein injection.Every group were further divided into three subgroups which was described as SCI 3d ?SCI 7d and SCI 14 d after injury respectively. The rats of every subgroup were killed respectively after assessment of BBB score. The immunohistochemical and fluorescent quantitative PCR methods were used to observe the pathological alteration of spinal cord and the expression of NGF.Result:(1)There was no neurological symptoms in rats of group A,while complete paraplegia neurological symptoms appeared in all rats of group B, C, D,the models of SD rats spinal cord injury was succesfully established.(2) there was no significant difference of BBB score within group B,C,D after SCI at 3d, BBB scores of each group increased in various degrees after SCI at 7d and 14 d,BBB scores of group C,D was higher than that of group B,BBB scores of group D was higher than that group C.(3)The significant expression of NGF m RNA and NGF could be observed in the injured spine, and peak around the time of 7d;the expression of NGF m RNA and NGF of group C,D increased higher than that of group B,the expression of NGF m RNA and NGF of group D increased higher than that of group C.Conclusion:The therapy of ganglioside combine with glucocorticoids pluse could promote the expression of NGF in SD rats synergisticly and then facilitate recovery of nerve function.
