As human living and eating environment becomes more and more complex, a variety of carcinogenic factors emerge which results in the increasing of the incidence of cancer. Tumor has become the greatest threat to human health and the tumor treatment has also become the focus of medical research. Chinese medicine plays an important role in cancer prevention and treatment due to the advantages as follows: stabilizing lesions, improving life quality of patiens, prolong the survival and no durg resistance. Anti-cancer Chinese medicine injection redeems the shortage of traditional Chinese tremendously. It can play efficacy better and is particularly important for critical patients. Human serum albumin(HSA) is the carrier for the storage and transportation of many endogenous and exogenous substances in the plasma, which can also interact with many drugs. The study of the interaction of anti-cancer Chinese medicine injection and human serum albumin is significant for the storage and transportation of Chinese medicine injection in human body, the clinical effect of drugs and the new grugs development.In this paper, the interaction between four kinds of anti-cancer traditional Chinese medicine injection and human serum albumin has been measured by the fluorescence spectroscopy, respectively. It is able to draw the mechanism of the interaction, relative binding constants, interaction force and the conformational change of HSA by analyzing the fluorescence spectroscopy and synchronous fluorescence spectroscopy combined with related theories.The specific research content of this paper includes the following three aspects. Firstly, the interaction between four kinds of anti-cancer traditional Chinese medicine injection(cinobufacini, shenmai, astragalus, brucea) and human serum albumin has been measured by the fluorescence spectroscopy, respectively. The results indicate that the four kinds of drugs can all quench the fluorescence of HSA and the quench mechanism is dynamic quenching. Meanwhile, the binding constants at different temperatures were calculated, and the binding force were mainly hydrophobic force. Secondly, the effect of each drug on the conformation of HSA was also analyzed by using synchronous fluorescence spectroscopy. The results showed that cinobufacini and astragalus barely affected the microenvironment of HSA. On the contrary, the hydrophobicity of HSA increased with the adding of shenmai and brucea. What's more, the internal hydrophobic structure became closer and the expansion degree of peptide chain increased, which indicated that the combination of the two drugs and HSA were more stable. Finally, comparison and analysis of the interaction between the four kinds of anti-cancer traditional Chinese medicine injection and human serum albumin had been conducted from quenching mechanism, binding constant, binding force and so on, respectively. The analysis results revealed that the four drugs exhibited differences in their affinities towards to HSA. And the order of the binding strength is: brucea > shenmai > astragalus > cinobufacini. All the information concluded from the experiments illustrate that human serum albumin can combine with the molecule of anti-cancer Chinese medicine injection effecitively to achieve transport and is significant for the clinical effect of drugs and the new drugs development.
