Objective:Using a two stage spiral flow channel microfluidic chip to sorting and enriching MCF-7 cells in the sample and compare the results with using stage spiral flow channel microfluidic chip once.Methods:Lable the MCF-7 cells with fluorescence dye. Configure the suspension of blood cells and MCF-7 cells at the ratio of 1000:1 (concentration of blood cells:108/mL, concentration of MCF-7 cells: 105mL). Using a two stage spiral flow microfluidic chip to sort and enrich the MCF-7 cells in the suspension at the flow rate of 400?L/min and then collect the sample of middle outflow tract. Using a spiral flow microfluidic chip to sort the sample for once same as above. Remove red blood cells in both samples by glacial acetic acid, count the number of cells of both samples and count the ratio of fluorescent cells in the sample of middle outflow tract using flow cytometer. Compare the results with each other.Result:When the concentration of cells is 108/mL and at the flow rate of 400?L/min, the spiral flow channel microfluidic chip can isolate cells well. When sorting and enriching for once, the chip can remove 91.51% of blood cells. When using the two stage microfluidic chip, the chip can remain a high capture rate of MCF-7 cells, and can remove 98.11% of whole blood cells.Conclusion:The spiral flow channel microfluidic chip has good ability to sort and enrich the MCF-7 cells in the sample.When using the two stage chip for sorting and enriching MCF-7 cells, it can remain a high capture rate and remove more blood cells.
