Objective:Using a spiral flow channel microfluidic chip to enrich circulating tumor cells of breast cancer for the optimum flow rate and separation effect.Methods:The qualitative observation of the test solution of different concentrations of red blood cells (106/mL,107/mL,108/mL,109/mL) at different flow rates (100μL/min,200μL/ nin,300μL/min,400μL/min,500μL/min,600μL/min,700μL/min) in the spiral flow channel microfluidic cell sorting effect determined the amount of its maximum load cells and the flow rate range; The quantitative analysis of blood cells inside of the flow channel recovery determined the optimal flow rate when the test solution of maximum cell mass concentration was tested at the flow rate within the range observed; We simulated circulating tumor cells of breast cancer using MCF-7 cells. Based on the maximum carrying cells capacity of the flow channel, the separation effect was observed when the test solution of blood cells and fluorescence preparation staining of MCF-7 cells (concentration ratio of 1000:1) was tested in the spiral flow channel microfluidic at the optimum flow rate, then the sorted cells were further cultured.Result:The maximum cell concentration was 108/mL when the spiral flow channel microfluidic chip isolated cells. When testing the solution with a concentration of 108/mL of blood cells at a flow rate of 400μL/min, the separation of the cells was the best. When MCF-7 cells were mixed with the diluted blood to be tested (concentration of blood cells:MCF-7 cell concentration= 108:105) at the flow rate of 400μL/min, the separation effect was good, and the sorted cells were alive.Conclusion:The spiral flow channel microfluidic chip has good ability of separating and enriching MCF-7cells, which simulates circulating tumor cells of breast cancer in blood, and little damage to the sorted cells, which provides experimental evidence for further circulating tumor cells research.
