Objective By comparing the contents of TNF-? expression between different groups,different times,under different drug intervention in DVT rats,plasma,explore the changes and significance of inflammation-related factors in the process of venous thrombosis;By comparing the contents of plasma tissue-type plasminogen activator(t-PA)and plasminogen activator inhibitor(PAI),differences in ET-1 levels and histopathology between the different groups,verify the relevance of corresponding Inferior vena thrombosis among each drug intervention group,to explore the role and the related mechanisms may exist of the herbaceous peony licorice Decoction during the treatment of deep venous thrombosis.Methods The experimental rats were randomly divided into two groups namely: the shame operation group and operation group.Using modified Reyers method prepare SD rat model of deep venous thromboembolism,to observe and record rats' general state machine and other local swelling behavior at each time point between the sham operation group and operation group,and randomly select two rats from each group for tissue morphologyobservation after 4h.The rats of operation group were randomly divided namely: The control group,Danshen tablets group,Herbaceous Peony Licorice Decoction low dose group,Herbaceous Peony Licorice Decoction middle dose group,Herbaceous Peony Licorice Decoction high dose group.along with the sham group,each group was lavaged in the different drug intervention,and detect the content of rat serum TNF-? by ELISA assay espectively,the content of t-PA,PAI by ELISA assay and the serum content of ET-1 by radioimmunoassay test;Take out thevenous thrombosis paragraph from each group rats after 14 days to observe the morphological changes by microscope.Results1.Changes in the rats at different times of TNF-?The content of TNF-? surgical group was significantly higher than the sham group compared with the control group 72 h after modeling.Herbaceous Peony Licorice Decoction low,medium and high dose groups were able to significantly reduce the secretion of TNF-?,and had significant differences(P <0.05,P <0.01,P <0.01);Danshen tablets group compared with the control group,the difference was not statistically significant(P> 0.05).The content of TNF-? 7d operated rats compared with sham-operated group was still significantly increased compared with the control group,Herbaceous Peony Licorice Decoction low,medium and high dose groups were able to significantly reduce the secretion of TNF-?,and had a significant difference(P <0.05,P <0.01,P <0.01),Danshen tablets compared with the control group,the difference was not statistically significant(P> 0.05).2.Changes of t-PA and PAI levels in rats of each groupCompared with the control group 14 days after modeling,each drug intervention group can improve t-PA levels,Danshen tablets group,Herbaceous Peony Licorice Decoction low,medium and high dose groups were significantly different(P <0.05,P <0.05,P <0.01,P <0.01).Compared with the control group 14 days after modeling,each drug intervention group can reduce the level of PAI,Danshen tablets group,Herbaceous Peony Licorice Decoction low,medium and high dose groups were significantly different(P <0.05,P <0.05,P <0.01,P <0.01).We concluded that Herbaceous Peony Licorice Decoction and Danshen tablets can increase t-PA,and reduce the role of PAI-1 level.3.Changes of ET-1 levels in rats of each groupHerbaceous Peony Licorice Decoction low,medium and high dose group ET-1 levels were significantly lower than the control group,statistically the differences were statistically significant(P <0.05,P <0.01,P <0.01);Compared with the control group,Danshen tablets groups was not statistically significant(P> 0.05).Herbaceous Peony Licorice Decoction medium and high dose groups were more significant.By Observing venous thrombosis formation section histological changes of each operation group rats,We found Recanalization phenomenon of early in the whole Drug intervention groups,of which Recanalization phenomenon in Herbaceous Peony Licorice Decoction medium and high dose groups were the most obvious.Thrombus wad dissolved mostly,endothelial cell was repaired and regenerated,wall inflammatory cell infiltration was light.4.Morphological changes of rats in each groupConclusion1,Herbaceous Peony Licorice Decoction can reduce the content of plasma TNF-a DVT in rats,reduce local and systemic inflammatory response in rat DVT,prevent inflammation further activation of the coagulation,and play the role of anti-thrombosis.2,Herbaceous Peony Licorice Decoction can improve the rat plasma t-PA secretion of venous thrombosis model,reduce the secretion of PAI,enhance fibrinolysis,which is conducive to absorption of thrombolysis.3,Herbaceous Peony Licorice Decoction can reduce DVT rat ET-1 synthesis,reduce endothelial cell damage,to play the role of antithrombotic effect.
