Regulation Of Soufeng Xiaoanyu Decoction In The Rat Model Of Bronchial Asthma In The Stem Cell Factor And Its Receptor (SCF/c-kit) Expression

Objective: To observe the relationship between Stem cell factor and its receptor with bronchial asthma,Discuss Soufeng Xiaoanyu decoction curative effect of treating bronchial asthma.Methods: Reference method,to determine about 6 weeks of 120 grade clean SD male rats as experimental objects,divided into 6 groups according to random number method,SXH,SXM,SXL,DXM,AM and BC.Put SD rats in simulated diurnal variation(12 hours during the day,12 hours of the night)of indoor cleaning,around temperature 20 ~26 ?,humidity 50~70%,all the rats raised a week after the experiment.Soufeng Xiaoanyu decoction groups,AM and DXM rats with egg protein 1 day 100 mg,100 mg aluminum hydroxide plus saline and 1 ml mixture,1 celiac injection of sensitization,the rats in the sensitization state;15 days of the ultrasonic atomization inhalation 2% egg protein every day,every time 20 minutes,1 times a day,to speed up breathing,cyanosis,labiali rats abdominal muscle spasm,nod breathing and unstable performance for the model set up a sign of success,for 3 weeks,to keep the rats of asthma symptoms.BC intraperitoneal injection and atomization adopt amount of saline.30 min before each sensitization atomization inspire began to fill the stomach,Soufeng Xiaoanyu decoction groups respectively with different concentrations of Soufeng Xiaoanyu decoction concentrate isochoric lavage,DXM with dexamethasone injection and lavage,AM and BC to the physiological saline lavage,a total of 21 days.Experimental 36 days take blood and lung tissue,and finally testing the following indicators: 1.Collect groups of rats serum,stem cell factor(SCF)level is measured by ELISA method,serum total IgE level detection,2.Based on each level upper left lung ? bronchial peripheral lung tissue specimens,tissue section,HE stain,line detection,optical microscope to observe and compare the different groups of lung tissue inflammatory change degree,3.Groups of left lower lung specimen,extracting total RNA,Real Time PCR technique to detect lung stem cell factor(mSCF/sSCF)and its receptor(c-kit)m RNA transcription level,4.Collect each upper right lung ? bronchial peripheral lung tissue samples,using Western Blot method,the detection of lung tissue membrane in combination with stem cell factor(mSCF)and soluble stem cell factor(sSCF)protein expression level.Results:1.Each group rats serum concentrations of SCF comparison: compared with BC,SXL,SXM,SXH,DXM and AM SCF density have different degree rise,with statistical significance(p < 0.05),in which SXH,SXM and DXM compared with no statistical significance(p > 0.05),SXL compared with SXH,SXM was statistically significant(p < 0.05),DXM and SXH,SXM compared with no statistical significance(p > 0.05).As shown in table 1.1-1.4.2.Each group rats serum IgE concentrations: compared with the BC,serum IgE levels significantly increased in AM,with statistical significance(p < 0.05),Soufeng Xiaoanyu decoction medium and high dose group,DXM,lower than the AM was statistically significant(p < 0.05),Soufeng Xiaoanyu decoction medium and high dose group and DXM compared with no statistical significance(p > 0.05),SXL compared with SXM,SXH was statistically significant(p < 0.05).As shown in table 2.1-2.4.3.The lung tissue pathology shows that: the BC lung tissue pathology images have no obvious inflammatory cells infiltration,alveolar walls are clear and complete structure without congestion,bronchial wall smooth,neat cilia;AM have more inflammatory infiltrates,lung congestion is obvious,wall thickening is obvious,the cilia fall off,the phenomenon such as adhesion,lodging;Soufeng Xiaoanyu decoction groups and DXM pulmonary inflammatory infiltrates the asthma model,SXM,SXH and DXM is most obvious.4.Lung tissue mSCF/sSCF and c-kit m RNA transcription level: compared with the blank control group,with statistical significance(p < 0.05),SXH,SXM,compared with DXM,no statistical significance(p > 0.05).Figure 4.1 to 4.3.5.MSCF,sSCF protein expression level: the rest of the group compared with blank control group,with statistical significance(p < 0.05),SXH,SXM,compared with DXM,no statistical significance(p > 0.05).Figure 5.1 to 5.2.Conclusion: 1.The SCF in serum of Asthma model rats and total IgE level is closely related to the severity of bronchial asthma.2.Different concentrations of searching the wind away dark blood stasis can improve the bronchus asthma airway remodeling in rats,reduce inflammation,and medium and high dose effect is better.3.sSCF,mSCF and c-kit With the incidence of asthma are more closely linked.
