Objective This study using compound diphenoxylate irrigation stomach prepared by slow transmission constipation(STC)rat model,and pass the Tong Mi mixture by gavage,changes of rat colonic ganglion cell of Cajal interstitial cells(ICC)to pass Tong Mi mixture in treating STC mechanism.Methods Healthy female SD 40 rats were randomly divided into control group,model group,mosapride group,Tong Mi mixture group,10 rats in each group.Using compound diphenoxylate(10mg/kg·d)model of STC rats by gavage method.Every day,a record of weight rats defecation and fecal wet weight,for 14 days.And Mosapride group and Tong Mi mixture group given corresponding drugs by gavage for each day and record a body weight of rats,rat feces number,fecal wet weight.Feeding 14 days later the rats fasted but not water for 24 hours.In the second day with self-made semi solid carbon paste(10ml/kg)intragastric administration,30 minutes later,the rats were killed,separation of rat colon,taken from the anal about 5cm in colon tissue about 1cm cut,was made into tissue sections.HE staining rat colon God ganglion cell,with immune group of labeled rat colonic ICC.Observed in colon of rats ganglion cell number and structure change;by image analysis software in colon of rats ICC area determination,through the comparison of the numerical difference in each group,indirectly observed changes in the quantity of ICC in the colon of rats in each group.Results Rats in the STC group were significantly decreased(P<0.05)in the daily feces and water content in the blank group(P<0.05).The group was significantly decreased compared with the control group(STC).Area of model group rats colonic ganglion cells and the number of c-kit positive cells were compared with the control group decreased(P < 0.05);mosapride group and area of Tong Mi mixture group rat colonic ganglion cells number and c-kit positive cells compared with the model group increased(P < 0.05).Conclusion Tong Mi mixture by improving the quantity and structure of the STC rat colonic ganglion cell of Cajal interstitial cells to improve the STC rats then quality,improve the moisture content of the feces,improving STC rat colonic propulsive function,so as to achieve the purpose of treating STC.
