| Objective To observe the changes of pathology and biochemical of the rats after human umbilical cord mesenchymal stem cells (HUMSC) by intramuscular injection.Methods A total of60male SPF Wistar rats were divided into six groups randomly:the normal group(PBS as the suspend liquid of human umbilical cord mesenchymal stem cells was injected in muscle); the control group with culture solution (named the culture solution group:DMEM as the culture solution of human umbilical cord mesenchymal stem cells was injected in muscle);the supernatant group of human umbilical cord mesenchymal stem cells (named supernatant group, the supernatant of human umbilical cord mesenchymal stem cells was injected in muscle);the low concentration group of human umbilical cord mesenchymal stem cells (named low concentration group:human umbilical cord mesenchymal stem cells with the amount of0.25X105was injected in muscle);the moderate concentration group of human umbilical cord mesenchymal stem cells (named moderate concentration group:human umbilical cord mesenchymal stem cells with the amount of1.0X105was injected in muscle),;the high concentration group of human umbilical cord mesenchymal stem cells(named high cell concentration group: human umbilical cord mesenchymal stem cells with the amount of4.0×105was injected in muscle). Each group included10rats and each rat was injected0.8ml liquid in muscle,0.2ml in limb for two times. The rat weight and biochemical tests were took before and after injecting the cells. The rats’life habit was observed during the experiments. Finally, all the rats were killed and HE stain was done with the liver, left ventricular wall, spleen, lung, kidney, brain and muscle. The ultrastructure of apex cordis cells was observed by transmission election microscope and the surplus myocardial tissue was weighted as wet and dry station.Results1The biochemical tests of all the rats were in the normal range before and after the intramuscular injecting HUMSC. There was no significantly statistical difference of the b(?)chemical tests among every group before and after intramuscular injection(p>0.05). Compared with the control group, there was no significantly statistical difference of the biochemical tests among the other groups (p>0.05).2The wet and dry heart weight of all the rats was in the normal range after the injecting HUMSC. After injection, there was no significantly statistical difference of the wet and dry weight of the heart and the total weight of the rats among each group (p>0.05). Compared with the control group, there was no significantly statistical difference of wet and dry weight of the heart, neither the weight of the rats between the other groups.3The HE staining of all organs and the ultrastructure of apex cordis cells of all rats were normal. After injection, there was no significantly difference of the tissues HE staining and the ultrastructure of apex cordis cells among every group. Compared with the control group, there was no significantly difference of the HE staining and the ultrastructure of apex cordis cells among the other groups.4All the rats had no symptoms of molt, skin ulcer, tachypnea, anorexia, diarrhea, and the rats had no death during the experments.Conclusions1No significant alteration was observed on HE stains of the tissue of the liver, the spleen, the heart, the lung, the kidney, the brain, the muscle of the limb after the injection of the HUMSC; minewhile the ultrastructure of the myocardial cell of the apex cordis was no changing.2No significant alteration was observed on the liver function, the kidney function, and the myocardial enzymes after the injection of HUMSC.3No significant alteration was observed on the weight and the living habit after the injection of the HUMSC. |
